    instead of the acquarium heater . you can also use a plant seedling heating mat + a PID temperature controller . vivosun has the mat + temp controller for like 25$. i happen to have all this i grow superhot peppers as a hobby . so if you do not have fish this maybe a option with dual use .

    Hi Gordo!, quick question, I have multi spore syringes, & was wondering what the best course of action to duplicate them would be?, should I just transfer to agar & isolate, then inoculate to grain to get a spore print?, or just use the isolated agar to make liquid culture? Much thanks

    • @bruhmomente7324
      @bruhmomente7324 ปีที่แล้ว

      Also, would CVG with calcium hydroxide work for panaeolus cyanescens, and do they require special agar, or would malt agar work?. furthermore will a liquid culture comprised of 500ml purified distilled water, 20gs 1:1 dextrose, golden malt, & 5gs peptone be a good liquid culture mix for panaeolus cyanescens?

    • @gordotek
      @gordotek  ปีที่แล้ว +1

      You can either direct inject to sterile grains or put the spores to agar, let it grow out, then transfer that to grains. Then apply the grains to substrate and grow it out to obtain more spores from mature fruit bodies. You can also use liquid cultures if you want, I don't like LC personally, easily contaminates and you can't always see that it is contaminated.

    Thanks for sharing such an informative video! Out of curiosity, is it a bad idea to wash the spores off of the foil instead of scraping it off with the scalpel?

    • @gordotek
      @gordotek  ปีที่แล้ว +1

      You can do it that way or even form the foil print into a cup and add water directly to the print, swirling and scraping it up with the syringe needle if you want (don't poke through though!)

    • @vincentwu2848
      @vincentwu2848 ปีที่แล้ว

      @@gordotek Oooh thanks for the advice, I've never done prints before so I have no idea what to expect. I figured if its powder on a sheet of foil, squirting water from a syringe would wash it right off but based on your feedback it sounds like it will take some agitation with the needle to get the spores off. I use 2oz condiment cups for agar, do you think it would be a good idea to try to take spore prints with a condiment cup instead of foil? Then I can place a lid on the cup after the spores drop, and then inject water through the lid and draw up spore solution..?

    Question,I'm having trouble understanding or answering by watching videos...... after you boil your grain, strain then sterilize, is the grain completely dry when you put it in jars and add your spores,or is it wet?

    • @gordotek
      @gordotek  6 หลายเดือนก่อน +2

      So, the grain itself absorbs water, it will look all "puffed up", there is lots of water inside, but not that much water on the outside. It shouldn't look very wet, for example there shouldn't be a puddle of water on the bottom of the jar. When you inject the spore water that will provide just enough extra water to get the spores germinating.

My question is this: Will it be successful after colonizing my jar (possibly, I would hope: ) then directly transfer the colonized substrate to the fruiting chamber and add only - first layer of perlite, middle layer of mycelium, upper layer of vemiculite. Are these nutrients enough? thanks in advance

      If the print was prepared in front of a flow hood, its likely very clean, I rarely see contams coming from prints although its possible. If the person who made the print didn't use a flow hood, its very likely full of contamination.

    • @gordotek
      @gordotek  5 หลายเดือนก่อน +1

      You can get away with as little as 0.5cc of spore solution if you shake it up good and ensure there are spores flying around just before injecting, but most of the time you want to do at least 2 or 3 injections so that would be 1 to 1.5cc minimum per jar (doesn't matter the jar size). You should see growth in the jar after putting in incubation temps for about 3-5 days, once it colonizes about 1/3 of the jar you can shake it up and spread the mycelium everywhere, then it will rapidly colonize all the grain (only do the shake up one time).

    May I please chime in regarding your laminar flow hood, so I noticed you have the inoculation set to Hight correctly within the correct parameters of your laminar flow except for one thing is that you have your inoculation site on a upside down Pyrex plate, which does not allow proper air pass through through the bottom and underneath of the Pyrex plate , you have placed upside down, which may result in a upward draft of your laminar flow. may I suggest a wired basket turned upside down instead of using a solid glass Pyrex, which may introduce a upward draft of turbulent air from the bottom part of your laminar flow setup remember turbulence is always your worst enemy when it comes to laminar flow I also noticed you have Foil that has been stuffed between the filter itself and your inoculation site on the upside down Pyrex plate to reduce this updraft so it is apparent that you are attempting to address this to a certain extent, but just to be on the safe side, I would rather use a wire basket and adhesive glued to a piece of glass or even some acrylic or something that is flat that you can glue on top of the wire, upside down basket to create a solid platform to work on and to remain free of worry of any updraft because of the wire basket will allow air pass through and even better no need to worry about stuffing the foil between the pyrex plate and your filter anymore🙃🙂🙃😎

    • @gordotek
      @gordotek  6 หลายเดือนก่อน +1

      You are very observant, and absolutely correct! I actually have a separate video on building your own laminar flow box where I do a better job, and use a proper wire rack, haha. But that laminar flow box in this vid is a massive commercial unit that is 2x4 feet in size, the sheer size and flowrate pretty much ensure no matter how silly my platform is, its unlikely to have a problem. But still, you are correct, I should be a better example.

      Only certain cultivars will do that, I am not a fan of this type of grow as the sclerotia are slow to grow and "dirty" when harvested, usually harder to grow, just seems annoying to me with no particular benefit other than some weird legal loophole in the Netherlands where sclerotia get special protected legal status for inexplicable reasons, haha.

Cantelmo was a sad story of mental illness, some actually think he was murdered but I doubt that. The craziest thing about his story is that he had been trying to contact the FBI about a conspiracy to infect the world with a dangerous pathogen, this was literally just a couple of months BEFORE the covid pandemic began! Ah well, wish he could have found the mental health help he needed. For what its worth, I advised him to STOP USING ALL psychedelics and to seek some help but I could not get through to him. He was clearly abusing DMT which I do not condone in any way.
