    You can really tell that he is coming from the ground, building this up. All the og tips that we get from him, if we don’t have the proper equipment are very helpfull. Today i just started my mykology class in university and i‘m so excited to do all the experiments like i see in you’re videos. Couple months ago i saw a video from you and that got me hooked up to mushrooms, so thanks a lot!

      What are you working with? Flowhoods are great, but it really isn't strictly necessary for small scale growing.

      There are definitely in between options... i got a complete HEPA laminar setup and coupled it with a 2x4 foot shelf to make a big work space... i was willing to spend a bit more, but under 1000 dollars for the shelving unit and the complete fan filter and hood setup. If you want to find a cheap HEPA filter go check out Staples and you could easily get a 9x9inch filter for about 30 bux that you could use to make your own clean air box. Question is just how big of a space you want to work in and how much you want to spend. I was lost in my early research but realized there are options for every price point

      @@SirCheese710 I wouldn't recommend using those HEPA air purifiers found in stores as a flowhood, generally their filters are not thick enough to create laminar air flow. IIRC, you need the filter element to be at least 6 inches deep for laminar air flow.
      I'd suggest going the DIY route if you are low on funds and a flowhood is really necessary ( it usually isn't). A 2x4 foot flowhood can be made for around $600

    I haven't watched a lot of your content yet, just found you recently. Very interesting content indeed and your stacked plate work was very good, I haven't worked in a micro lab for over 15 years so lovely to see. You may cover this in other videos bit in case you don't, your refering to aseptic technique when you talk about maintaining sterility in a lab, I thought that might help some people who want to research further about it. (For the you, it's not important, as you clearly know what you're doing).
    Also for others the plates come in the bag, sealed for sterility, if you open carefully it perfect to then store them in again, with a little masking tape to close up. I'd only store them like that, if storing blank plates (Petri dish with set agar) before use. I'd tend to use cages to store inoculated plates, but they lived in climate controlled cabinets.
    Also interesting fact about agar for those new to it, you heat it up to about 85°C (about 185°F) to liquify it and it will then stay a liquid as it cooos to around 37°C (about 100°F) then it gradually set.

      Actually the laminar flow hood, blows air horizontally or vertically unidirectionally. In this case it's horizontally towards him, and away from the cultures. Kind of like a fan, and doesn't get recycled.

      You can see that there’s a though process to each and every process, yes there might be a reason for each process, and logically here might be a better process. However, for some reason we stick to the same process, and that’s usually because it works best (most efficiently for the goal set)

    Really great vid! Just what i needed to see. And yes it’s a bit intimidating with al the sterile stuff as a beginner. I really want to start my own little mushroom farm. And this helped a lot! Exited to see more

      You can certainly do it! My wife and I started a mushroom farm over the past year. It is challenging, especially if you encounter a lot of trial and error, but this is something worth it and very rewarding.

      @@vishnu2407 We grow mostly indoors, but some outdoors. We live in South Louisiana, U.S., so its challenging to grow outside in our hot summers.
      Our lab (clean room) is 10 x 16 ft. And our fruiting space is the same. So that's 320 sq. ft. total, however it would be more helpful to have around 1000 sq. ft. with a dedicated space for colonization.

    Thanks man! Great set up and simple explanations. One question: Why is liquid culture inoculatation not good for substrate (e.g. sawdust substrate)?

      I'm a year late but screw it, nobody's answered and you've got a few upvotes. So the mycelium in the culture is forming very loose networks in the nutrient solution that are easily broken apart when transferring, but with grain, it has a solid source of nutrients to form a concrete network on so it will have a much easier time branching out and colonizing a new substrate from there. It's like if you dropped people randomly into a forest they would eventually meet up and pool resources and could form a society eventually but they would have a much better chance if they were dropped in as groups that are already working together and well supplied.
      Continuing with the analogy, these groups of people would also have a better chance of surviving a lone wolf attack than a lone person would against a pack of wolves, so you're gonna want to thin out the predators next. With grain you can work in small batches, it is nutrient-rich and you can sterilize them so your mycelium has no competition and can easily become established. With bulk substrates, pasteurization is commonly used by beginners because A) it's difficult to properly sterilize large quantities of a substance without a large (and expensive), industrial autoclave and B) pasteurization allows a few organisms to remain to keep each other in check so one doesn't get the upper hand over the other. Then when you drop in well-established networks of mycelium special forces can easily overpower the scattered wolves or nematodes or Trichoderma or whatever and take over the place
      Edit: There are other reasons for the culture to grain, grain to bulk substrate process too. For example, some species will not fruit on grain and need a wood diet in order to reproduce. Other species, like cordyceps sinensis, only grow on insects. So why are you putting them in grain spawn? That won't work. You just look silly

    Got a love those gorilla grow tents. I got three myself and I wouldn’t buy any other brand. watched a friend brake a tent Because the plastic frame. I that will have them for years.
