    Best advice I have ever seen on you tube. "Put your LC to agar before you use it". I am a huge fan of LC but hes right, you really do have to monitor your LC.

    You will need: 2 Mason Jars, 1 with a normal metal lid and gasket, one with filter and injection port. Coffee filter. 600mL warm filtered water. 1 gram of ELME (extra light malt extract) or LME. Spore syringe.
    1. Add hot water to mason jar. Dissolve 1 gram LME to water.
    2. Place metal top on upside down (so it doesn’t seal), and screw on gasket.
    3. Pressure cook for 20 minutes at 15 PSI.
    4. Once depressurized and jar is cool enough to handle, secure coffee filter on gasket of another mason jar and strain LME solution.
    5. Remove filter, replace with lid that has injection port and filter.
    6. Cover lid with foil and pressure sterilize for 45 minutes at 15 PSI.
    7. Once depressurized, let cool for 12-24 hours.
    8. Inoculate with spore syringe.

    I honestly absolutely despise whenever I see a comment on your channel about the facts that you take too long to get to a subject or over explain. When I'm coming to your channel to get as much in-depth knowledge as I possibly can. Every second of your time that you take to give me this video I cherish thank you Willy and these people that are hating on you for that kind of thing keep it to yourself. What was the golden rule you learned in elementary school? Every long winded explanation of his is absolutely packed full of real knowledge not just some random guy in a forum. The knowledge this man spreads is absolutely expectacular. Either way mush you all. The fact that you're on this video speaks for itself.

    Hi Willey, your content is wonderful, but you are a rock star from the voice to hand the gestures, positivity, and breakdown of everything. I'm 58 and who says you can't teach an old dog new tricks. Energy level through your videos has a great tempo like a fine song. I just found out that it was due to Mushroom microbiology that the smallpox vaccination was created. The Mushrooms potential is truly miraculous. I don't have pets but now I feel like I do. It's incredible to view life on a micro level and thanks to you a new world has been opened up. keep up the good work.

    Love your content man. Made a smaller version of your dub tub incubator, and it works like a charm. I added a $20 humidifier that runs off a water bottle I'm put it inside and I've never seen grow so quick I almost don't even have to water. About to have my first fruits thanks to your guidance man. Just really trying to understand the science of it and how to prevent contamination or errors other than sanitation reasons.

    The clones I've made, are doing beautifully in this culture following this recipe. I did 300ml water with 1/2 gram of the extra light malt extract powder. I think more research is needed on how much energy spores, tissue samples for growing out and mycelium liquid culture expansions really need in the short term. The standard recipes call for much more sugar than this, but I also hear of people keeping their liquid cultures in a cabinet for many months and they are still viable. It's amazing how much we still just don't know about fungi.

      So 0.5 grams LME per 300 ml’s of H2O = 0.17% same ratio as Mr. Willy.
      12 grams is 4% of 300

    Just startihg out..Found your site online and got some spore sringes from you. I gotta say, from not knowing a thing about anything regarding shrooming..... I've got an awsome start because of your site and others. Yes, it is an awsome community and just this fact has really been a huge help with the anxiety and depression I've been trying to get a handle on for most of my life! Thanks for all the love WM! The small bonus suprise that came with the order is really a big thing (thanks!😊) and says a lot about what you are about. Looks like I will be getting more involved with mycology. The spores are now turing into 3 all-in-one bags of beautiful looking - and feeling, cakes. I can't wait to get to the next step of trasfering them into their new home.     1g per 600mL is no where near enough carbon for a healthy culture. it's important to have a proper carbon/nitrogen ratio. around 3-4% sugar solution is fine. that's 3-4g per 100mL water (600mL = 24g LME/KARO/Honey/Dextrose/Sugar or any combination of these have proven very successful for me.

      @@alexwalston5469 Thanks for that. Its been a while since i wrote that comment, and i've experimented a lot. For spore germination in LC (which is not generally recommended since spores are inherently dirty) 1-2% LME is about right since it'll enable the mycelium to branch out looking for resources. However, in terms of a healthy LC, let's say it's for for an isolate, one would be looking for 2-4% LME in my experiencce. Thanks for the reply.

      I tried several different recipes, some still working, some cultures I had to throw out. All my cultures using 1gram are still good. I don't know the exact reasons but whenever I use more than 1 gram, the culture eventually gets contaminated.

      I use 5g LME + 0.5g Nutritive yeast + 250ml water for smaller batches (basically my agar recipe) Is there a reason Willy uses such a tiny amount of food ?
      As for the contam talk, either your culture is clean or it is not. LCs are not for any use other than perfectly clean tissue samples imho, otherwise that sample should be going to agar to continue cleaning up the culture

    my favorite recipe for liquid nutrients is:
    8 grams of corn syrup for every 100 ml of water. (PC for 30 mins at 15 PSI)
    it's dirt cheap & your mycelium will love it.

      Should be pressure cooking a little longer than that. That may be long enough to sterilize the liquid but not the emdoporses embedded in the vessel. Just saying.

    Have a question for you. Just purchased an amazing book 'cooking with magic mushrooms' it's written by a Dr and seems a chef was also involved. Some amazing recipes dude, especially jelly sweets and hard candy etc.. Including toffee like consistency. Great stuff on chocolate too and how to get dosing down to exact amount.
    Would love to hear you views on cubensis extract/tincture?

    Hey Buddy........4% of 600ml Water is 24 Gramm, in the case of 400ml it is 16 Gramm..............1ml Water weighs 1 Gramm

      If you added 24g to 600ml water then the total volume would turn into 624ml. 24 might be 4% of 600, but it won't be if you add it to the 600ml volume.
      It needs to be 4% of 600. Not add 4% to 600.

    I would say the honey and water is probably the best there are books from the 50,s that mention honey and water cultures. It's finding the best mix for the mushroom your growing honey by it,s self has antibacterial properties so just making sure you use good clean water is enough most of the time. 🍄🍄🍄

    1 gram of LME in 600 mls of water is much, much less than 4%. It's actually 0.17 %. (1 gram sugar in 600 grams of water).

    I pour my 1gram ELME into 500-600ml boiling water from kettle, and just quick stir with spoon.. it liquefies in seconds. Been doing this for years.

      So a couple weeks ago I read your comment and thought oh yeah now I’m not gonna bother with the straining and all of that. I should’ve just listen to the video instead of listening to your comment Bc the issue is that even when you use boiling water and it liquefies after you pressure cook it there’s still going to be a bunch of sediment and your liquid culture is going to look disgusting, it’s also going to be hard to see anything you put in there.

    Just a polite FYI - 4% of 600ml of H2O by weight is 24g, 1g is appx. 0.017% of 600ml by weight.

      In science related matters it’s important to state objective facts… Math is the way, the truth, and the light!
      • 1 ml of H2O = 1 gram. Fact
      • 4% in decimal notation is 0.04 or 4/100
      Therefore:
      • 600ml of H2O = 600 grams
      • 4% of 600 ml’s or grams is written as y=0.04(600) = 24 grams
      So if the maximum is 4% by weight the range of sucrose would be from 0 - 24 grams per 600 ml’s of H2O.
      Accuracy is paramount!

    Willy_myco, I have a different procedure for l.c. that works great and will save you a little more time. Just a little different..I use a pot 1st, add my filtered water, add my gram of light malt, then I add .5 gr yeast nutrient. Then I bring it to just boiling and remove it from heat. If I need antibiotics, I add that after I remove the pot from the heat, allowing it to cool a little 1st. Next, I don't flip my lids..I want them to seal to keep out contamination. I only pressure cook for 15 min at 15 psi. If I'm isolating wild mushrooms, or colonizing a dried specimen, I use the antibiotics to isolate the mycelium and make sure no hitchhikers are present from being handled or from bugs, etc. The yeast nutrient causes a faster, healthier bloom. If the l.c. isn't clear in the beginning, it will clear in 3-5 days as the nutrients are ingested. I was not expecting the yeast nutrient to make that much difference, but it did. Everyone, buy a magnetic stirrer. It speeds up the growth and is only $40 on Amazon. I have porcini, shitake, white button, plus more in the fridge right now. You are most definitely correct, light malt is better than other sugars...I used karo, then honey, and they worked, but not as fast or as efficient. Now you guys can work on isolating and selecting genetics and achieve faster results. Try a little yeast nutrient in your agar, too. You should get roping faster and heavier.
    The amount may need an adjustment...I ran four 1/2 pint small mouth jelly jars and they seemed to get exponential growth quickly...enough that I told you about it..lol..wes tek...
    Another thing that will affect your l.c. grow time is temperature. Know what the specimen prefers if possible..but pretty much all do well at 72-75°with low to no light...I have run my l.c. under a halogen desk lamp 12 on 12 off as well, and they grew fine.

      hey Wes , i saw you said Agar ... so you use the n/yeast in agar too ? i just picked up some to use in agar but now ill try it in l/c . i used to be worried that cause my l/c wasnt clear clear it was nfg , but ive found it does clear up as the culture grows . i thought i was caramelizing it .why is 4 % of 600 ml 24 grams not 1 gram ? do you have teks out there under Wes Tek ? Thanks for any info. you can share . i use a sab but still getting 50 % contambras w/ my agar , l/c is going good . An We Be Chubby ova' he'ah'

      @@MostWantedOHA nothing out there for wes tek...just thought since I haven't seen anyone else using yeast nutrient, I would name it. Yes. It will clear, and you don't have to heat it, but it mixes better if you do. Gentamicin will help with contamination if you use it in the beginning..only necessary for wild isolation or if you're cloning a dried specimen that has been touched..I have dipped specimens in alcohol before inoculation and that works too if you can't get gentamicin. There's other antibiotics you can use too, like erythromycin..you don't want to go over 4% sugar...a little over won't kill it. If you were using honey, 24 gr would be correct. If you're using light malt extract, it only takes 1 gram. Idk why, just following the tek. I experimented with the yeast nutrient and found it very successful and any mycelium I have introduced to it thrived faster and looks very healthy..some even floated to the top and started growing, with black spots in the middle that appear to be trying to pin. That was a dry albino specimen I found. Next, I will add y.n. to my agar dishes to see how it works. I expect the same results. It's not necessary, but if it affects the growth in a positive way, then it's a bit better tek. I wanted to put my name on it since I had a successful experiment and no one else has named it that I've seen...just for acknowledgement.
      If you're having contamination issues in your tubs, add hydrogen peroxide to your misting bottle or tank if you have an auto setup. Also, wear a mask and use gloves. People say it's not necessary, but trust me, if you want your mycelium healthy, contaminate free fruits, and more flushes, then don't breathe on it or touch it. Just like a loaf of bread, it will start mold just from your touch or breath.
      I have an extract method too that is awesome. I have not released it yet. I'm about to start a few medicinal grows, & I will shoot the method when I have a flush of rieshi or something to demonstrate with.

Thanks Wes ...i recently tried enthrmicin , they use it in fish tanks it aint cheap either , im good w/ spawning my tubs got 7 flushes from one last year . my contams are coming from just the air in here got central a/c . its only when i open anything to outside o2 that im getting the dreaded tric ...im sure its in here since ive been growing for a year now , gonna step up w/ a laminar flow hood , been shopping for filters an collecting info. i sprayed down some g.t. w/ iso an talk about blu , wow . im having better luck w/ h2o2 doing clones in l/c and agar . the hydrogen peroxide really made a big difference in success rate . you ever check out home mycology , aka mycomadness ...

      Just trying to check measurements how much water do you mix with your .5 gram yeast nutrient and 1gram lite malt extract? Is it a half quart of distilled water? I was going to use this solution to my spawned seed culture once it’s fully colonized then do just a quick mix in the blender then pour that on a layer of rye seeds that will be mixed with my various substrates. Thoughts?

    Let the LC sit for a few days before you inoculate it. This will allow you to verify that no contamination made it through the PC process.

    Hey Willy, I could go from a slant to LC right? Would probably take a little piece of the slant and make a liquid inoculant and then inoculate the LC solution. Does that sound right? Or would you take a piece of the slant and inoculate the LC solution that way? It would skip a step which I'm a fan of but don't know how to inoculate the LC without taking the lid off of it but I'd probably have to take the lid off of something somewhere anyways. If I were using a normal agar plate I'd just suck up some LC solution and spray the plate and suck it back up but I don't want to use an entire slant doing that.

    So I can take a tissue sample from a t3 and open the lid and scrape the tissue into it and the the LC will grow that way?

    I'm extremely new to this, but I follow directions well and have the necessary equipment minus the agar. How do I know what the LC is supposed to look like? I can't tell the difference from mold. Any sites with pictures? Thanks in advance

    You should just microwave the plain water in the lidless jar, add your malt, stir up, filter, and then pressure sterilize? Save a ton of time and energy.

    There are so many videos and forums on making LME LC out there, nut this one is really clear and explanatory. Thanks Willy!!
    One thing I don't understand though is the water/LME proportion used here. 1g LME to 600ml water is actually .1666% (1/6), which is 1/24th or the 4% ratio Willy himself mentioned here. It seems minuscule!
    So is there such a large range fo LME to work that it would go from .16666 to 4.0??
    Any clarification would be very appreciated ;)

    So what did he add to the sterilized malt extract and water? Did he inject directly from a spore syringe to create the liquid culture? Or did he take a piece of an agar plate and put it in the sterilized LME water? I didn't see that anywhere in the video.

    Love Willy but he makes a short story long and at least in this case, makes a somewhat simple task somewhat difficult.
    Throw 650-700 ml ow water in a sauce pan, bring to simmer, throw in LME stir and filter to LC jar.
    Then you PC with some grains or by it's lonesome and inoculate. Should be gravy in a week or so.

      Grains are generally PC'd for 2hrs @ 15 PSI. When sterilizing agar or LC its generally PC'd for 20-30mins @ 15 PSI.

    Very helpful video . But did I miss something or was the jar that had the coffee filter as well as the filter itself not sterilzed? I find it hard to assume the liquid is hot enough to kill everything in the jar especially in the top airspace and lid change.

    In my spore syringes (they are in a 0.9 % solution of sodium chloride) there is like some green stuff in the base of the interior of the syringe, but when I look it closely, it disappears. Is it contamination? The thing I don’t get is why I would have a contamination in just salt and water

    Could one use this liquid culture mixture as the liquid additive to turn ones inoculated grain spawn into a grain spawn slurry instead of distilled water? Would it help it grow faster when poured and mixed with your substrate?

    Yo willy do we even really need a pressure cooker tho if ur using lids with a syringe filter or flipping lids so the jars dont build pressure cant all this be done just by scalding the jars in a regular pot of boiling water from preparing grain and LC and substait whats the point of a pressure cooker when were not actually canning for the purpose of preservation if Steralization is all thats required

      I add 1 gram soy peptone and 1 gram extra light malt extract to all my 600ml jars I make on a fullscale mushroom farm. always works fine.

    willys such a G, i had that same problem. karo lc with tons of growth but anytime i spray some lc into some grain it never would do anything 8 out of 10 times. very weird. i dont even knows hows that possible.

    Is there any difference between Light malt extract (LME) and dry malt extract (DME)? In my country I can only find the latter

      DME is Dry Malt Extract. that's what us home brewers usually call the stuff. There is also a version that is made with wheat, rye, and there are darker versions to.... LME refers to the color, personally I would prefer if they just called it Pale rather than light. I bet you would be fine using any one of the renditions of DME on the market... I actually plan to test that soon.

    I had micropore tape on my lids, I also put the aluminum foil on them. I did the second pressure cooker run for sterilization, and I ended up with LME water on the bottom of my pressure cooker. It seems there was some evaporation and the LC just came out of the jars and got mixed with the water on the bottom of the cooker. This must have changed the ratios in the jars. Anyone had a similar problem? Got any suggestions? I guess I could do the sterilization with regular jar lids, and also sterilize some lids with the injection ports and the micropore holes, maybe wrap them in foil as well so that I can open it in the SAB and change the lids in there.
