    The needle biopsy move is a good idea. Never thought of using the needle tip for a quick and easy way to get a piece of tissue

    Thanks for the comment about stirring releasing Co2 and allowing O2 in. The fish tank analogy helped. I have some liquid culture growing and I was stirring twice a day, I just put it on the stirrer and turned it on, and will leave it there.

    Thanks man. I used this technique for the first time with a still air box. 1 mushroom to 3 different jars. All 3 jars cleanly colonized :)

    Thanks for the video MCM. We use needle biopsies for most tissues. The mushroom is dry compared to human tissue but anything that is soft can be biopsied pretty easy.

    Excellent technique! I'm considering doing this with morels. Im going to try to go from a jar to a 5gal bucket. Then dump the bucket over a prepped burn site in the fall.

    I use a technique to clone mushrooms directly in the forest. I take some sterilized butyl cap vials filled with liquid medium and use only a needle with a cilinder inside, without the syringue, to push out the piece of mushroom tissue. From now I'll use this technique, as only needs the liquid in the vial to push out the tissue.

      hahah for real, i wanted to clean my computer back in july or so, and had to get alcohol swabs because the entire alcohol shelf was barren at every store... even the witch hazel was always gone!!

    Healthcare best practice for stetile technique is to wipe in a reverse spiral ... that is to start in the middle and spiral outward. I think it would be best to end by coming off of the table away from the fan.
    Might be overkill but its evidence based.

    That's so cool. Not messing around with spores and just using fresh shrooms. I've read that 0.3% hydrogen peroxide kills contaminations and spores but allows to grow the mycelium. However I have found no videos yet.

      Psychedelics are the reason why i didn’t take my life when i was at my end. I was stripped of my ego and saw the beauty of life and interconnectivity and even though i still battle anxiety and depression, I’m doing better everyday and will never think in such a self destructive way again.

      LSD and mushrooms completely changed my whole outlook on life. I became a better version of myself
      This experience gave me a lot of confidence about my self and my body. A bunch of bad thought / behavior patterns were broken. One of these was pretty bad OCD that made me wash my hands a lot. It gave me a lot of hope that things will be fine, this is the one thing that I heard throughout the trip: Everything is alright. The main reason for the trip was my severe depression and it definitely helped me (although it's not gone). Before all I could do was lay in bed. Now I am trying to rebuild my life one step at a time which wasn't possible before."

      Hello Mossy Creek mushrooms. I went to a location yesterday that was supposed to be monster mushroom company had to drive for a little while to get there I got there and the people were rude to me they did not have the thing that I wanted they did have some sterilized grain and some sterilized substrate so I bought a bag of sterilized substrate for $30 got home with it and come to find out there is a hole in it and where they tried to seal it there is a hole big enough for me to stick four fingers and and it goes straight down to the substrate so I did not get a bag of substrate that was sterilized I got a bag that was unsterilized really frustrated at this point and probably am going to be looking at what you guys have for sale.

    Great content - I like the idea of the needle biopsies better than going from tissue to agar and then liquid culture. Less opportunity for contamination by exposure. Question: What is the best needle gage to collect a proper sample? Also, at what temperature do you store your liquid cultures while they propagate? I am using 1/64th teaspoon of peptone, 1/64th teaspoon of light malt extract, and 1/2 tablespoon of corn syrup with 600 ml of hot distilled water. What is your favorite recipe?

    1) Put gloves on, spray gloves with alcohol
    2) spray table, spray jars, spray the floor and the air for funsies until I flee slightly light headed from all the iso in the air
    3)Spray my spray bottle with separate iso sprayer
    4) Spray my hands again, spray the jars again
    5) Spray the table one more time for good measure
    6) Innoculate a jar or two
    7) Spray the shit out of the table, and my hands again
    8)Flood lab with 500 gallons liquid death
    9) Drain lab
    9)Continue work as per usual
    Few questions
    How often do LC jars need to sit on the magnetic stir plate? How long can they sit without being stirred?
    Looks like the grey lids you're using are the new "leak proof" lids for mason jars. Just curious what are your thoughts on plastic mason lids vs metal? Rust aint fun to deal with but even Roger Rabit on the forums at the shroomery holds the opinion that plastic lids contaminate more. Seems unlikely you would be using them so much if that was your experience. Thoughts?

      Once a day is good enough for stirring with magnetic stir bar. More would be better. As for lids plastic works just fine. Just tighten again after autoclave as they sometime get loose. The whole contam thing has prolly more to do with the filter disc getting too wet and bacteria getting in that way.

    How long should you stir for? Also once the culture is established, how long will it last on the shelf? How do you store it after the culture is established?

    If someone is really OCD about sterilization they can put items under UV-C lamps as well as pressure sterilization ... I use them for my breathing masks and filters ...

    Wow. I have always gone fruit to agar to second agar to liquid culture. Good to see this is possible. I will try direct to LC. I have some LC jars cooling right now. It is my first time using the grey leak proof lids. So far, so good.

    How long do you recommend each stirring session (if done daily) should be, if using a magnetic stir plate? 5min.? 15min.?
    Btw, this is definitely my favorite cultivation channel as a total newbie. You're extremely thorough and give fantastic explanations, actually describing *why* things are done, rather than just saying "do this" because it's what others say to do.
    Plus, I love Norse culture too (I see the Mjölnir necklace and the Icelandic magical stave on the door) so that's a bonus🤘🏻 Anyway, thanks for all your videos!!!

    Thanks dude, I totally learned a new technique from you today! Thanks for sharing. I also purchased some liquid culture pre-mix (and used your code) from shroomsupply. I will give LC another go because my first try using Karo corn syrup was great at growing mycelium and mold no matter how clean my process was using a SAB. I also noticed when using your flow hood you should elevate your work surface just a bit so its not right at the very bottom of the flow hood. Anyway great vid, can't wait to try this out.

      here's a few suggestions to try
      pick the best type - there are many available.
      grow them in the best environment - some eg oyster muchrooms like cool humid conditions and may be placed outside (I read these and the reasons they work from Gregs Mushroom Grower site )

      As a microbiologist/geneticist I can tell you that this is NOT a problem. You are not making a copy of a copy. The analogy that would be more fitting is you are taking a cutting from a tree that was grown from a cutting.

      @@michaelgrouse7850 oh that makes more sense. So it's more like propagation rather then a clone thing. Ya when i think of clones i think of the star gate series with the Magvyer guy. The one alien species was like dying off cause they couldn't reproduce so they cloned themselves and was like dying off. Haha it's good there's real geneticists out there to counter hollywood and the vividness dilemma.

    My liquid culture solution is easy peezy. Gummy Bear TEC. I use 3% gummy bear by weight = roughly 1 bear per each 100 ml water. Organic Black Forest Bears are my preferred brand. There is a little gelatin in there as a protein source. The lower ph from acid and juice might reduce contamination?

      Can you give me more info? Someone gave me a bunch of those gummies, and I want to use them before they turn funky, and that looks to be a nice, practical use. Do I just dissolve the little boogers in my water?

      @@melhawk6284 Start with a ball jar with a liquid culture lid (and a stir bar if you have a magnetic stirrer). Add 3 grams gummy per every 100 grams water. Pressure cook the jar for 30 min at 15 psi. Cool. Inoculate. Stir the jar regularly while culture grows.

    Hey bud.. I just wanted to say a couple of things. First.. yalls lab is BAD ASS! I am quite quite envious. I intend to be building one soon as well. Wont be as big as yours but.. if all goes right and the Good Lord Jesus wills it so. its gonna turn some heads :) If and when I build it, I may make a video to show you. And lastly.. I dont think you use too much alcohol at all actually. I think it is perfect. I plan to do it exaclyyy how you do it with the alcohol. Better to have a bunch of nothing go wrong, rather than use a little and all hell breaks lose. Keep up the good work bud! :)

      @@vinodpunj3102 I'm not expert but I would probably say no. If it's completely dry it probably doesn't have any actual living tissue to multiply itself from.

      If you guys are still wondering, it does work. I ran the idea to a friend who has a certain interest in mycology. He gave me a few dehydrated (probably with cool air) samples that he had sitting around and forgot about for over a year and a half. I didn't have a needle at the time, but I used an exacto knife that was wiped down with alcohol and flame sterilized to carve a small chunk out, after exposing the center with the same process splitting it open like in this video. The other thing I did different was adding it to an agar recipe I learned instead of to liquid culture. Did a few more times in case of any possible contamination from the first, which I did have a couple contams, but with a lot more that were successful. Not sure if mushrooms dehydrated with heat would have any possibilities as well, or mycorrhizal varieties that usually need a plant host to survive. Will probably run tests on those theories eventually, but for now I am just excited with the success I had with this. And I hope it helps others out too.

      My understanding, and I'm a complete noob, is a dark warm spot. I've seen 70-80 F is a good range
