  • All the details you need to know on how to make a liquid mushroom culture of a gourmet oyster mushroom, although this technique can be used for 100's of types of mushrooms.
Liquid culture consists of a sterilized nutritious solution that has been inoculated with fungus spores or mycelium.
Andrew goes over each step from sanitization and to shows you exactly how to take a sample directly from the mushroom, put it into the liquid solution and how to care for the culture as it grows.

The needle biopsy move is a good idea. Never thought of using the needle tip for a quick and easy way to get a piece of tissue

Thanks for the video MCM. We use needle biopsies for most tissues. The mushroom is dry compared to human tissue but anything that is soft can be biopsied pretty easy.

Thanks for the comment about stirring releasing Co2 and allowing O2 in. The fish tank analogy helped. I have some liquid culture growing and I was stirring twice a day, I just put it on the stirrer and turned it on, and will leave it there.

In labs liquid media is the preferred method for culturing, simple Erlenmeyer flasks are stoppered with Cotton wool plugs and then wrapped in tinfoil to make sure the wool does not get wet in the autoclave. Color changing Autoclave tape is stuck to the bottle to indicate the bottles that got sterilized. You culture a generation one or more master mediums and then fill the flasks from these. You can also inoculate from a petri dish, mushroom or another liquid culture by first sterilizing an inoculation loop in a flame and then transferring your subject (save on syringes). For agitation we use a lab shaker vs introducing a stirrer mainly as you can shake multiple flasks continuously while incubating them. Consider a flask and cotton wool to save $ and an inoculating loop over syringes, unless your selling them. Also consider experimenting with say a 2-4% Glucose solution for media. If you really want to get crazy look at methods like pulsed electric field sterilization of bulk substratre or growing mushrooms continually in liquid on raft scaffolds like hydroponics but for mushrooms - fungiponics :D Looking forward to your next video.

    • @arrhazes8198
      @arrhazes8198 3 ปีที่แล้ว +4

True. For the Stock Culture we use a slant agar- slower growth, lasts longer.
Then transfer to petri dish agar as Mother Culture (using inoculating loops).
Then from petri dish to liquid broth in hundreds of flasks on shakers (using inoculating pipes/borer-don't remember the exact term). Lesser liquid in larger volume flasks results in much higher growth speed but means more flasks and shakers needed.
For really large scale we used Fermenters instead of shakers.
I think we never used a single syringe for production. We use glass or metals apparatuses that can be autoclaved and reused indefinitely. Single use stuffs are for the hobbyists not professional production.
      Just sharing..

Hello Mossy Creek mushrooms. I went to a location yesterday that was supposed to be monster mushroom company had to drive for a little while to get there I got there and the people were rude to me they did not have the thing that I wanted they did have some sterilized grain and some sterilized substrate so I bought a bag of sterilized substrate for $30 got home with it and come to find out there is a hole in it and where they tried to seal it there is a hole big enough for me to stick four fingers and and it goes straight down to the substrate so I did not get a bag of substrate that was sterilized I got a bag that was unsterilized really frustrated at this point and probably am going to be looking at what you guys have for sale.

Thanks dude, I totally learned a new technique from you today! Thanks for sharing. I also purchased some liquid culture pre-mix (and used your code) from shroomsupply. I will give LC another go because my first try using Karo corn syrup was great at growing mycelium and mold no matter how clean my process was using a SAB. I also noticed when using your flow hood you should elevate your work surface just a bit so its not right at the very bottom of the flow hood. Anyway great vid, can't wait to try this out.

I use a technique to clone mushrooms directly in the forest. I take some sterilized butyl cap vials filled with liquid medium and use only a needle with a cilinder inside, without the syringue, to push out the piece of mushroom tissue. From now I'll use this technique, as only needs the liquid in the vial to push out the tissue.

Hi Andrew. First, thank you for your YT channel. I'm new to home growing of mushrooms and have found your videos very helpful and full of great information! Secondly, I will be using your discount over at Shroom Supply, so thank you for that as well! My question regarding liquid culture is this: how long should I place my culture jars on the magnetic stir plate each day? I'm starting working with only two types of gourmet mushrooms (oyster and lion's mane), and I've already ordered culture syringes for both types, but once the rest of my supplies arrive and I inoculate the prepped/new culture jars, what length of time will I need to let each jar sit on the stir plate? Thank you in advance for any advice/guidance. And Happy Thanksgiving to you and the entire Mossy Creek team! Peace & Respect.

Healthcare best practice for stetile technique is to wipe in a reverse spiral ... that is to start in the middle and spiral outward. I think it would be best to end by coming off of the table away from the fan.
Might be overkill but its evidence based.
