    EDIT: The tubing inner diameter is 1/4" and the outer diameter is 3/8" (for the master LC lids)

      Same... I was searching everywhere for the purpose of the syringe filters and nobody talked about it. So I would ask in the comments and nobody would give an explanation. Found this channel and this guy has already answered 3 of my questions in 2 videos.

    I made diy lids with silicone and micropore tape, works wonders, but i never thought about how the hole is needed for air displacement. Great insight!

    @ 10:23 That obnoxious bend in the tubing ends in the middle of the jar so you'd essentially only be able to draw half of your LC through the tubing. You could get the last half with a syringe but that defies the whole point of using the tubing imo

      The length was for a quart jar, & I'm sure he was just using the half quarts as a demonstration.

      yes and also if you tip the jar sideways the tubing goes to the side and you can draw the remainder that way - it’s not perfect but this was an early version

    How do the mycology lids tubing not get contaminated after pressure cooking?

      if it’s already connected and capped off it will be sterile until it’s opened and then it’s a matter of doing proper aseptic technique

    Small question:- while the culture is being preserved in fridge, is it necessary to do fresh transfers every 2-3 months as they run out of nutrients??

      I've had a culture of Nepal chitwan that made it in room temperature for two years so in the fridge it should be fine for at least longer than 3 months

      @@louisw5858 it's taking a s syringe and sucking some mycelium out of one liquid culture and then innoculating another jar of fresh liquid culture such as honey water or corn syrup water with the syringe.

      You can transfer to be safe and that's better but I don't think it's necessary as often as one might think

    How do you keep the end of the piping sterile? Especially if you cap it off with a syringe or leur lock cap wouldn't any lc remain in the end section? Wouldn't it be preferrable for it to run back into the jar? Thanks.

    Excellent content, as usual. Have you got an update on the morcella garden?
    Also, which glass pint or quart jars work best with the magnetic stirrer? I bought a case from my local garden center and they have a fairly exaggerated concaved bottom which sucks because I basically used all my culture on them to learn that the magnetic stirrer will not work with them

      thanks for following along! I just used microsoft word and imovie with my iphone - keep it simple and easy! 🙏🏻 I have an Imac for video editing and my macbook for all the thumbnails, titles etc. People on Fiverr helped me a lot in the beginning and then it took a lot
      of practice to get honed in. Some of my earlier ones are still up and quite different then the style now - MUSHLOVE

    I'm sure your aware of the mono tub tek, and flushes. How do you feel about using grain water for misting and rehydration for flush?

    does the pipe get contaminated while storing? since we're using the same pipe to connect syringes multiple times ?

      this is the caveat - I keep a bunch of spare tubing sterilized in pouches and if I suspect this, I just swap for fresh tubing. The key is to never have the liquid go back to the main container that way there is a break point you can just swap between pulls

    Hey. I had a question about the advanced lid with the clip to prevent back flow.
    1. How do you extract a full syringe of LC when there is air in the tube? When I try it only fills about 4cc
    2. When you snap the clip to close the tubing, how do you expel the liquid in between the syringe and clip to prevent air in the system and prepare for subsequent LC extractions?

      you can either ad a backflow preventer or just keep the hose kinked between syringes. Also, I will leave the last syringe in use on the end as a “label” and as a cap to keep air from entering the system. Things have calmed down a bit here I want to make a recap video soon

      I would love to see I recapping vid. I bought the master Lc lids and gave the jar with the lid bacteria. Im not sure where I’m the process I mess up but I’m sure it’s user error on my end.

      they are reusable to a certain extent. Once they get discolored I swap them (after a few runs)

    The one with the clamp...doesn't solution get stuck between the clamp and the tip where you insert the syringe, and then spill out when you pull the syringe off?

      no, there isn’t any back pressure the clamp acts as a backflow valve

    Gary, some of the parts on your list seem to only have a temp rating of 175F. Have you found this to not be a problem?

      it shouldn’t matter unless it’s tubing, don’t use vinyl tubing but what I have listed is what Ive been using and it works great

    Thanks I needed this. My idea (what I'm presently using) of the early days is the original jar lids I modified with hobby lobby felt and silicone. I'm assuming all of the components of these lids are vulcanized right?

    I make my own. And for my lids. One 5/32 and one 7/16 drill bits are perfect sizes for my filter and injection ports. No adhesive, just a very tight press in seal. I just pull out the filter and SHIP when I think they need replaced. It's so easy to do It doesn't feel right selling them.

      yes I have done this and am planning a system to scale it, there is risk with contamination but it helps grow it fast

      no foil needed just be careful to not pull them by the tubing and it works well

    you do realize the back of those syringes are open and exposed and then you aspirate back and forth you are introducing contaminates right. 8:37

      Yes that is the whole point to displace the air while aspirating and filter it through .2 microns did you even watch the video?

    The problem with putting a syringe on immediately after inoculation is that the majority of the syringe is exposed to open air until you decide to take your first sample.
