Pro tip for anyone doing this for the first time, always run your drill in reverse when drilling plastics to prevent cracking.
@Valerian Roots Yup, if you're drilling plastic, no matter the size of the hole, run the drill in reverse and don't push to hard. Works every time.
My guy you are my hero. I’m starting up my company and trying to get all aspects for sale, including culture lids. Went to mass produce them and about had a stroke from how frustrated I was getting from the lids cracking. You are a god.
Pro tip. Use a one of those triangle stone bits and drill through like you would a step bit. Its just sand through so there is no risk of craking or breaking at all
Or use a drill & brad point bits...
I don't understand this comment. With no special effort, I've never cracked a plastic mason jar by drilling. Frankly, if I was doing a shotload (ahem) of these, I'd probably pop for a drill press and do 10 at a time.

I love the way you hold the jar to get good angles on the pieces, you can see the growth off tot the side of the first little lump of mush
If you forgot the aluminum foil also I found a really good way of keeping the coffee filters over the jars by using the original ball rim the The jar came with Nice deep and snug

Thank you so much for the footage of the development. I put Lion's mane LC in such a jar to let it grow. After a week I only see a tiny bit. But I asked smart people and they said it is alive. But way slower than the life you showed there. Good luck with your work!!!

I bought the HaBeuniver 20mm injection ports, the shorter grey ones that seem to be very common, and they'll fall right out of a 1/2" hole but a 7/16" is a perfect fit. Thought this might save someone buying the wrong bit. A brad point bit works great on the standard metal lids, btw. Great video, Ashley, as always.

You don't need an injection port. put two layers of foil on the top of your jar. Lift up the first layer and inject culture or spore into the jar. It's been working fine for me for years.
Any other tips for anewbie? Spores n all in 1 grow bag should be here this week
@@trump45and2zig-zags make a still air box. Its hard to explain, you get a tote and make two holes for your hand and get some pvc pipe or something your hand can fit through. Glue some of those big rubber cleaning glove to the inside and seal everything up. I put glade wrap over the top of mine so I can see better. Put everything you need in, and spray some rubbing alcohol in, then put the wrap over it and let it sit a while.

10:45 it's also the perfect time to 'noc up a few more jars with that LC so you can save some time on turnaround. Instead of having to go from the tissue biopsy stage next time. Lovely video BTW! Makes me want to get growing again. So much faster than cannabis with zero electricity required and basically one brief moment of light just to start pinning? You can't beat that. I'm just super weary about trying to go myco-psycho again because the last go around I got arrested for unrelated reasons and wasn't able to return home for a few weeks. By that time I had 8 Rubbermaid tubs that were just an orgiastic death feast of aspergillus and black mold plus some contams I'd never even seen before. Ever since then I can't even keep brand new bread in this freaking apartment for 2 days without it molding. I'm terrified if I put new tubs in they'd have the same exact fate.
By doing the tissue biopsy you select the best genetics from the mycelium. A pity to hear what happend. Anyways if you esterilize properly Im sure your not going to have any problem.
You're going to need fungicide spray. Look it up, set up more than one in your case. Perhaps everywhere you put tubs in the past as well as one for the common areas. Set the sprayer up and leave the apartment for the day. Come back and you should be in better shape.
Amazon has those lids in colors, with silicone gaskets. They are sweet. I use 1/4 drill grey injection ports, and 1/2" stick on air exchange glued down with red high temp RTV sealer. They work great!

Is that an injection Port filter? I have a question about injection ports when you tip the jar over doesn't the filter get wet?

@@BoomerShroomer I ordered five, the first was bad. I lost one bag, and the spores. I have four left, and there is no way I will try them. That is $50 down the tubes.
@@jameskreiter how do the other ones look? As long as they smell good and you can’t see contamination they should be good to use. I use his bags lot and rarely have issues.

Have you used peptone When you make your liquid culture it’s introduced 1 g a very beginning it has amino acids in proteins that will make a forest of mycelium in liquid culture it’s pretty amazing. Thanks for your previous help it worked out really good for me thank you.

I took morel culture possibly correctly, the matter that has formed is the same shade of blue Grey and the bases I plucked from the ground. I put the container outside with some holes in it bc it's suppose to be cool and moist today

Hi Ashley I have a newbie question. What temp do you leave your liquid culture for colonization and is it in complete darkness the whole time? And when the jar is fully colonized, how long can it be stored for?

Do not use blunt tip, they will cause too much damage to self-healing inserts. Hypodermic needles are available from mycology supply stores online.

@@wanderleybecerra2763 If you have a still air box or a flow hood, go for it. I just transferred some lion's mane and blue oyster from agar to LC that way a week ago as I'd run out of liquid gasket.

Start to give it fresh air and fan it for 30 seconds twice a day. That will start pinning.
If you're using hydrophobic GE filters you don't need to use foil for your lids. Leaves you plenty for spore prints! Mush love Boomer Shroomer!

Spore syringes and LC syringes are different. LC inoculates much quicker, also. Spores take longer.

Liquid culture is not spores it is live mycelium
Yes a syringe of culture will work but spores need to germinate into mycelium so it has to go to agar/mycelium/ then to a LC

I’m brand new to this. Built a still air box, I have a colonized spawn bag coming. I’ll be making my own substrate. I have everything for agar plates, slants, and am wondering would it be asinine to also have LC? I’m growing lions mane as my first go.
First video I’ve seen that shows the initial growth, I looked for hours finding a video that didn’t show a week later when it was all cloudy and chunky and I thought my white clumps was mold
Nice! It is hard to see contamination in LC. It is always best to test on agar.

The liquid culture might take the the paddy straw but I am not 100% sure.
Here's a question what gauge and length are those needles where do you get those I've never seen those super long needles I've only ever used regular ones or long blunt ones but if you're going through a self healing port it's best to have something sharp or you're not getting through it

You do not need to keep the jar in a dark place just out of direct sunlight. Light does not affect the growth because they do not have chlorophyll.

Just makes it easier to stir and gets completely dissolved. Not necessary. Thank you!
Some people use a few small pieces of broken glass in the jar, (placed before pressure cooking) to help break up the mycelium, if they don't have a magnetic stirrer .

I want to echo everyone's thanks. These videos are excellent. Regarding breaking and shaking the bag, can you do this too often and if yes how many times is enough, 1,2,3...? Thanks
2 is more than enough, anything over that and you might risk killing the mycelium or putting it in stasis. I usually only need to shake once at around 35-50%
