    Hi there, please help! I am conducting the antibacterial assay of my plant sample following the well diffusion method. But I have faced with a problem. Should I filter my plant sample with the help of 0.2 micron filter inside laminar flow cabinet before adding the samples into the wells. I found that when I filtered my sample, the concentration as well as the volume of my prepared sample also get decreased. I have also gone through severe literatures of my field but found no paper mentioning the use of 0.2 micron filter. So is it necessary to do so or I can skip this step?

      No. You can do the test without filtering your sample. Though chance of contamination may remain, you can use 0.45 micron filter.

    Hi, Nice video. I am totally naive in this. I want to check the antibacterial activity of silver thin films and small scaffolds. Will you please guide me? Is it necessary to use Amoxycillin antibiotic disk along with our sample?

      Antibiotic disk is necessary to check the validity of the test result.
      But you can do this test without using antibiotics disk.

    Hi! I am still quite naive in this topic. I have two questions I wanted to ask :
    1. if I am testing an antifungal activity of a substance (garbage enzyme from citrus peels to be specific), can I use well diffusion method?
    2. do I inoculate the fungi in the same way as shown how to inoculate the bacteria in the video (by streaking the entire surface of the plate)? Or is there a better way to do it with fungi (ascomycete)?
    I am using Botrytis cinerea as the fungi btw.
    From what I can understand so far (by researching the topic on the internet), the standardized/more reccomended method for testing against fungi is to use broth dillution, but I am also under the impression that well diffusion can also work to test antifungal activity againts Botrytis cinerea, from a study I had found titled "A Comparative Study of Antimicrobial and Antioxidant Activities of Plant Essential Oils and Extracts as Candidate Ingredients for Edible Coatings to Control Decay in ‘Wonderful’ Pomegranate"

      You can check antifungal activity by disk agar diffusion method. Only difference is the inoculation system. Collect spore and dilute in saline water and inoculae into agar surface. Rest of the method are same. We will upload a new video for it. Stay with us.

      I used an antibiotic disk and a treated fabric sample. You can analyze any sample with antimicrobial properties

    Hey I'm working on a fabric treated with copper nanoparticles (turns out I actually synthesised copper *micro*particles but let's not talk about that). Do you think I can use this method to test the antibacterial activity? I've been searching for methods and was thinking between ATTCC 147 and JIS L 1092 but I've figured out my fabric samples were not fit with aatcc 147 and I couldn't find enough information about jis l 1902. I would appreciate some help :)

      You can use this method to check the antibacterial activity of any treated fabric sample. You can use any bacterial strain and there will be no problem with it.

    Hello! I've got a fabric treated with silver nanoparticles. Do I need to sterilize my samples before I test the antibacterial activity? If so, how do I sterlize them?

      No. Your sample already have the antimicrobial property. So no need to sterilize the sample. But during the development of nano particle, you should take measures to avoide much contamination.

    how about antifungal activity for seaweeds against m.fijiensis can be the same method? also the agar will be PDA

      No. Not same. Fungal spore suspension is preprepared and the rest will be the same. SDA, PDA or DRBC can be used.

    For thoes who don't know we can use either one Nutrient agar or muceller hinton agar or Tryptone soya agar for in the petri dish

      Use a sterile black disk and soak in you soap sample solution. Then using this same experiment you can analyze the antimicrobial activity.

      For powder sample, you can make a solution by dissolving your sample in a suitable solvent.
      Then prepare the MHA plate inoculating with bacterial.
      Now bore the media with the aid of sterile borer or something else and in this bore add 50 microliter sample solution. Then follow rest of the procedure according to this video.

    Maam, how much agar that we can make with 38g of MHA medium ? in the video, i only can see 1 complete agar plate? if i need to make about 8 agar plate, how much do i need to use? Thank you in advanced

      In the media presentation, you need 25ml liquid MHA solution for each plate. So if you want to prepare 8 plates then you need total 200ml liquid MHA solution.
      To prepare 200ml MHA solution, you need 7.6g MHA dehydrated powder (as 38g needed for 1000ml or 1L solution. So 7.6g needed for 200ml solution).

      1. Purchase sterile blank disk from Oxoid.
      2. Prepare streptomycin solution.
      3. Soak the blank disk into the streptomycin solution.
      3. Dry the disk in air.
