Touchdown PCR

  • Touchdown PCR - This lecture explains about the mechanism and principle of touchdown PCR reaction and the application of touchdown PCR in amplifying DNA fragments. Touchdown PCR is done to eliminate the formation of unwanted DNA amplification and it increases the specificity of the PCR reaction process. In touchdown PCR, we start with a very high annealing temperature which is close to the melting temperature of the primer and slowly reduce the temperature as PCR cycles are conducted. By this manner unwanted primer binding can be inhibited and specific PCR amplification of the target gene can be achieved.

    all you said is correct unless one point is the definition of the melting temperature , you said it means the temperature at which the primer get damaged . however in PCR there is a beginning temperature of 95 degree to separate the 2 stands of DNA which is very high and do not damage the primer . so the correct defenition of the melting temperature is that it is the temperature that nearly below it the primer can bind specifically to the DNA strand and if we go far away below it , the primer bind unspecifically ti unwanted regions of the DNA.

      No , but the melting temp of primer is where primers get deattached from the strand i guess

    in google and various other sites, it is mentioned that the annealing temp at starting is 5-10 degrees higher than tm not lower. You had used the wrong term IG

    TD-PCR purpose is to figure out which is the best annealing temperature?, and if that is correct, how do you know at which temperature the primers binded correctly?
