DNA Replication in Prokaryotes | Initiation

- In this video we have discussed the Initiation part of DNA replication in Prokaryotes where the enzymes and proteins attach to the DAR sequence and initiate the process of unwinding . The unwinding is done by Dna A protein and then other proteins accompany the initiation process.
The Proteins involved in process are :
Dna A protein : ADP and ATP Bound.
Dna B : The Helicase enzyme which breaks the Hydrogen bonds.
Dna C : Helicase Loader.
Dna G : Primase enzyme.

@@jyotisoni9814 Most probably it could be the Structural genes organisation ( structural genes include lac z lac y and lac A genes -operan ) there are several other structural genes but these are the most imp ones

Great explanation, Hussain but doesn't Ori-c sire in prokaryotes contain 9 nucleotide motifs of 5 repeats & not 4 repeats as you mention here?
I agree with you! There is for sure 5 repeats of 9-mer motif. Plus at the end of his lecture, he mentioned a DNA pol III holoenzyme, that in fact has three DNA polymerases (not just two!): one of them is working on the leading strand and another two on lagging strand - "trombone model".

My professor gave us a different approach as to how the dsDNA becomes ssDNA at the 13-mer region. They told us that the ssDNA-binding-domain in the DnaA Protein causes the strand separation and didn’t mention an introduction of topological stress due to the coiling of the DNA around the Protein DnaA.
