Always. I find that the fondant is so much easier to work with and is much smoother with the tylose added. It doesn't get rock hard (I don't add too much), so you can still cut into it with no problem.
You can do it a few days (up to a week) in advance if you want. But you will want to keep it sealed in a ziplock bag until you're ready to use it so it doesn't dry out!
