  • Using Chromatography to Investigate the Pigments Isolated from Leaves of Different Plants. Required A level Biology practical activity. Make sure you see the end where calculation of Rf values is explained. Method could be used to compare pigments in different plants such as shade loving and sunlight loving species.

    A small correction here though, at 8:15, the RF value must be smaller than one, not zero 👍

      Instead of alumina plate, one can use silica gel plate that can be made from silica gel added with solvent and poured on a glass plate, leveled and kept in oven.. silica gel plate ready to use now

      Hanna Bouftah it’s gritty texture just helps to break up the lead tissue.

      Nurarihyon hafiz it dissolves the phospholipid membrane so that more pigment is released and acts as a solvent for the pigment.

      The different pigments have different solubilities in the various components of the solvent. The mixture gives better separation.

      Sorry I can’t help with that. It was a while ago and I don’t have the raw results. The distances will change depending on the type of stationary phase and exact composition of solvent used.

      kaylia walker You could try it. You are unlikely to get a good separation but you should get something.

      @@africanqueen9820 Hi biology student here. From what I know, you can't use distilled water or even water to separate the pigments since the pigments are located in the thylakoid membrane inside the chloroplast, which means we are dealing with a membrane and membranes usually have a lipid constitution and if you want to 'break' the membrane to dissolve the pigments, you absolutely need an organic solvent, which is why you use acetone and petroleum ether. Please correct me if I am wrong.

      Theo You calculate it and compare to standard values for pure pigments.

      Theo there are a few txt books with them in or you can run pure pigments yourself with the same solvent .ill see if I can send you a link to them.

      Jasmine Wallace-Thompson if you use pen, the ink will dissolve in the solvent and run up the paper obscuring the results.

      Jasmine Wallace-Thompson if it wasn’t it would dissolve in the solvent rather than being carried up the paper by the solvent.
