Column chromatography devised on the basis of differential adsorbance(partition coefficient) of substances on solid adsorbent (eg:silica or alumina) to an extent, that depends on the substance polarity (considered for solubility) and other chemical properties and structural properties. Some compounds are adsorbed more strongly to the stationary phase than other and thus they will be eluted (washed down) the column at a very slow rate, thus can be separated on the basis of their elution rate accordingly. Thus naturally coloured components of spinach Spinacia oleracea) and other leaves can be separated by this technique. The dark colour of the spinach is a combination of β-carotene and chlorophylls.
