Needles are gamma sterilized, so rubbing alcohol and flame cannot improve sterilization. Those steps can be omitted. When injecting, rather than pulling the needle out and re-inserting 3-4 times at different angles, just insert the needle one time straight in, inject a tiny bit, then manipulate the needle from side to side without pulling out of the injection port. Pulling out and re-inserting risks contamination. Good video. How was your flush?
I know this is old but, maybe you can still help? I’ve been told to flame the needles because the heat will cause the injection port to heal back better? Is this bs?
@@kelseyevans8451 yeah, that’s bs. The injection port is self-healing. The flame does nothing to a sterile needle but leave carbon behind. I don’t know why people invent theories. Just ignorance I guess
That's all well and good for one bag. Replacing a needle for each and every jar or bag is obserd. Heat sterlization is effective and used when having to inject 100's of bags or jars.

When it comes to desinfection alcohol, make sure to read the manual on how much of it must be applied. In hospital we must soak everything so wet it takes about 5 minutes for it to dry, or else there is a chance bacteria has not died.

I appreciate the nice video with narration. Although there are a couple of things that seem odd for someone that did sterile environment work in college. -The flame sterilization of the needle doesn’t make sense unless you are concerned that you grain bag is already contaminated: the port, grain, and needle are clean. I think this is more important if you are cutting agar as you don’t want to transfer between plates or stratification for contamination. SIDE QUEST: the soot is sterile. -Spraying the needle is odd, as using good sterile technique will be more effective (placement of items, moving left to right, not breathing on the specimen, etc.)

Great vid! 1 suggestion, after flame sterilizing the needle, there's no need to whipe with alcohol. By doing so, you are basically downgrading the sterility, since you are going from sterilized to just sanitized.
What about the soot left over on the needle? If you don't wipe it with alcohol swab, then what about the black soot left on the needle after flaming it? Will that contaminate?
@Howard Hopkins just remember that there may be soot in your grain spawn. I forgot one time and thought there was contamination, but it was just the soot lol

I have probably watched this and many of your other videos 50 times each just because I want to make sure I do this absolutely correct, in preparation I've been monitoring areas in my apartment for the last 2 weeks as far as temperature goes and because I live in an old building and not all the windows are up to date it can get pretty cold, luckily it never drops below 55 in my apartment because I live in San Diego. My question and main concern is is that going to ruin any progress if the temperature drops below 70° for a few hours each day? Anxiously awaiting your reply as I just inoculated tonight
@@jocko1283 so funny story, this comment was about my second attempt because my first attempt on January 11th didn't seem to work and I thought I might have overheated my grain bags trying to counter the less then ideal temps I was dealing with. However, I was getting ready to toss the first attempt 3 days ago when I noticed it had some growth going on so they might be saved lol. As for the second attempt it's only been 7 days and so far no signs of mycelium. Fingers crossed though 🙂 feel free to message me directly if you want any more updates
@@jocko1283 yeah I have seen videos where it takes a week to see growth all the way to 45-60 days until growth so just wait a week before you even look at it again otherwise you'll end up like me checking every 10 minutes like something is going to happen haha where did you get your spore syringe?
Mushrooms grow best in a cool environment - but not too cold! 70 degrees Fahrenheit (21.1° Celsius) is an optimal temperature for colonization and fruiting. I typically keep a heater set at 70°F in the room, and this seems to yield good results during both stages. If you can't use a space heater and have to use a heating mat, I recommend placing a towel between the heating mat and your tub. Make sure not to set the heating pad too high, 68-70 degrees fahrenheit would be as high as I would go. If the temperature of the room in which you're growing drops below 65°F, then your mushrooms will grow very slowly, or possibly even stall. Avoid high heat as well: the mycelium has a death temperature of 104°F (40°C). Keep in mind that the temperature inside your monotub will typically be a few degrees warmer than the room it's in.
If you inject too much spore liquid into the bag it can lead to bacterial growth from the amount of moisture in the bag. You typically don’t want to inject more than one cc of liquid into each bag.
I like your videos and really appreciate your content. I don't mean to be a dick but there's just 1 thing I'd like to point out, liquid won't pour out of a syringe if you hold it pointing down, it would only occur if there was an air port at the top of the syringe allowing a pressure differential. I only say this as it makes sense to me to hold it pointing down to limit potential contamination for amateurs like me that only use a still air box. Look forward to more of your shared experience and wish you well!
Just had a big problem with these bags. Wish someone could come help me learn where I went wrong.but a little after doing everything you just did, some brown sugar type dust formed at the top not sure if it was the spores or the bag

One question please. I'm burning my mind on how to.know the pressure inside the pressure cooker. If its less than 15 bar it would not have the same efficiency. I have just a regular kitchen 5L pressure cooker. So again, how can I make sure everything is OK? Thank you legend
I am loving your videos. I’ve watch them all. Let me see if I get this right: if using a grain bag you start with inoculating the bag letting it sit for over a month in a dark warm spot. Then add to the substrate in a mono tub keeping the lid closed from 10 days or more - warm and humid - I noticed one of your tubes had a tube piped into it -is this necessary at this point Or can I run a humidifier nearby? Last lift the lid of the monotub for the next weeks to come as your spores fruit while Fanning and spritzing 3+ times a day? This is a complex project and requires patience and unfortunately a lot of the TH-cam videos are not as comprehensive as yours. I really appreciate the work you’re doing
Yes exactly!!!! You don’t need a humidifier or fan. You can do that yourself and the Monotub will keep the correct humidity itself. Feel free to reach out to me on Instagram and we can talk through the process once you get started. But you totally get it!
I had inoculated 96 Grain spawn bags with syringes from premium spores with exactly 3cc in each spawn (3pound bags) that were bought from Mycohaus. I turned off the ac injected the bags in a clean closed off area/ environment in my spare bathroom everything wiped down with 70% isopropyl alcohol along with spare closet in there then sprayed air with Lysol made sure everything was wiped down as in video. Temp was 75-80 in around march here in California spawn bags were white doing well took about 2-3 weeks for them to colonize 50% then broke them mixed them around waited about a week or 2 and they took longer then expect to fully colonize they all got about 70-80% some even 90% while waiting for all to be colonized they started to get “built up orange murky “water at the bottom of bags mushy type brown spawn towards the end did a little research to find out it shouldn’t smell sweet any reason why that happened?
When prepping the bag spread out the grains and mix in the bag they’re tightly packed and just came from sterilization, you need to introduce air into the bag first. Way to much liquid into the bag too just 2 cc’s per pound is plenty you can do 2-3 bags per 10cc syringe.
I went out of my way to actually make sure I ordered your actual mono tubs to support you.. I love your videos.. So I have a HUGE question ‼️ I just injected about spawn bags, and 1 all-in-one bag from mushroom supplies... AND I JUST SAW A VIDEO THEY SHOWED THAT SPRAYING THE PATCH WITH ALCOHOL WILL RUIN THE FIRST LAYER, AND CONTAMINATE IT🤬⁉️ Please tell me this isn't true. I don't understand why people would all stress about the alcohol WITHOUT mentioning to AVOID THE PATCH🤦🤦
I had no idea you had to shake the syringe that much. I shook it a lot and saw things floating around. I injected it all into a single place on my grow bag but nothing is showing 3 weeks later.

Where the most ideal places to keep spawn bag while waiting for the mycelium to grow? I have mine in a card board box w a temperature of 75-76. Is it ok if I have it in a open room? (Basement). Hardly any traffic down there. And when people say in a dark place does it literally have to be pitch dark or is it fine in my basement with lights turned off, and as I said before nobody comes down here
Is the mycelium always supposed to look like that? Does it matter what strain? I’m trying PE and there is definitely white stuff growing throughout the bag but it doesn’t look anything like that.
I did this and couldn't hear the hussing sound when i whiped the needle after burning it!!! 😂 i guess my earmuffs playing music made me lose on conforming the hissing sound of the needle cooling off after rubbing with alcohol .
Hi! I did everything correctly besides the fact I may have injected too many spores. I have a 2lb bag and injected around 10ml. Have I ruined it? It is a spore syringe.
The port should be self sealing but if you're worried clean the port after injection with alcohol and dab some silicone on hole. I use high temp sterile silicone based caulk. High temp is not needed though

I screwed up my filters seems like.. bags were so tight, airtight, had no air in them at all, didn't make any sense, so I removed white thin layer of the filter, then slowly the bags started to get air, which would make much more sense now. Well beginner here, lets see what happens..
@DaddyDank There could be many reason why you got mold... One your spawn bags could be contaminated even before you got them, your spores could be contaminated too.. or the environment could have had mold. Were you in a closed room with no air circulation? I recently inoculated 5 bags. Two were good and three got contaminated. I think the contamination came from the spores.

Starting my first grow this week thanks for video I heard when it’s 80 percent break it up some say 50 yours said 20 so iam a little confused but iam going to try one bag your way other bag 50 percent

I used premium spores to buy my spores. So far they have been very reliable. The best spawn bag I bought was from this guy. spawnmagic.com/?itm=BoomerShroomer Only thing was it took more than a month to get. I heard he was going through a hard time and thats why his shipping was delayed. I think they have better shipping times now. But so far these spawn bags are by far the best. I used some other ones that had mushy grains and smelled.. They still worked but would never fully colonize. I am also in the process of making my own spawn bags. I will have the video up in a few weeks. Hope this helps!

Don’t do that or don’t do that again. IME, growth took forever for me to see progress. It was probably growing within the middle. I also can’t tell if you’re using MSS but I was using LC. You do not have to listen to me but if it works for you, it works for you. Most people I’ve seen say it’s a bad idea because you really don’t want to mess with the grains while they try to settle. Instead. Try to inoculate in different points of the bag and make sure you’re really careful for this part, by injecting towards the screen of the bag so you can keep track of progress
My biggest concern is ordering syringes on line. Not for the spores or possible contaminants but it seems like a way for others to figure out where I live and what I'm doing. Is this concern unfounded?

Suggestions for maintaining the optimal inoculation temp? Do you use a small heater?

I ended up buying a pressure that had a gauge. That was the only way for me to know exactly what the pressure is.

I just injected about spawn bags, and 1 all-in-one bag from mushroom supplies... AND I JUST SAW A VIDEO THEY SHOWED THAT SPRAYING THE PATCH WITH ALCOHOL WILL RUIN THE FIRST LAYER, AND CONTAMINATE IT🤬⁉️

I had no idea you had to shake the syringe that much. I shook it a lot and saw things floating around. I injected it all into a single place on my grow bag but nothing is showing 3 weeks later.

Hi, so the air is not important in the the grain bag. I so some others allowing air to go inside the bag before sealing it using impulse sealer.
Yes, air flow is important. You do need some airflow for germination and colonization in the bag.

Where the most ideal places to keep spawn bag while waiting for the mycelium to grow? I have mine in a card board box w a temperature of 75-76. Is it ok if I have it in a open room? (Basement). Hardly any traffic down there. And when people say in a dark place does it literally have to be pitch dark or is it fine in my basement with lights turned off, and as I said before nobody comes down here

I did this and couldn't hear the hussing sound when i whiped the needle after burning it!!! 😂 i guess my earmuffs playing music made me lose on conforming the hissing sound of the needle cooling off after rubbing with alcohol .
I forgot to sterilize tje needle but never touched it when it came out the package is my bag contaminated now? Damn it 😪

Hi! I did everything correctly besides the fact I may have injected too many spores. I have a 2lb bag and injected around 10ml. Have I ruined it? It is a spore syringe.
No, it should be fine. Mix it around in the bag and then let it sit to grow. Wait 10 days to see if you have any growth.

I don’t.
The port should be self sealing but if you're worried clean the port after injection with alcohol and dab some silicone on hole. I use high temp sterile silicone based caulk. High temp is not needed though

Normally 2-5ccs. I've put the whole syringe in and it was fine.
I screwed up my filters seems like.. bags were so tight, airtight, had no air in them at all, didn't make any sense, so I removed white thin layer of the filter, then slowly the bags started to get air, which would make much more sense now. Well beginner here, lets see what happens..

You need air flow. Leave the filter up.

Love all your videos. I tried this. I must of done something wrong. I got green mold in my bag, guess will try again.
@DaddyDank There could be many reason why you got mold... One your spawn bags could be contaminated even before you got them, your spores could be contaminated too.. or the environment could have had mold. Were you in a closed room with no air circulation?
I recently inoculated 5 bags. Two were good and three got contaminated. I think the contamination came from the spores.

So far, his bags are the best that I have purchased so far. I don't think there is a local supplier for spawn bags. You will need to buy them online.

Starting my first grow this week thanks for video I heard when it’s 80 percent break it up some say 50 yours said 20 so iam a little confused but iam going to try one bag your way other bag 50 percent
I say earlier. When its about 20-30 percent. Break it up and mix it around and reset. It will colonize in half time. But it is not a prefect science.

Only thing was it took more than a month to get. I heard he was going through a hard time and thats why his shipping was delayed. I think they have better shipping times now. But so far these spawn bags are by far the best. I used some other ones that had mushy grains and smelled.. They still worked but would never fully colonize. I am also in the process of making my own spawn bags. I will have the video up in a few weeks. Hope this helps!

Don’t do that or don’t do that again. IME, growth took forever for me to see progress. It was probably growing within the middle. I also can’t tell if you’re using MSS but I was using LC.
You do not have to listen to me but if it works for you, it works for you. Most people I’ve seen say it’s a bad idea because you really don’t want to mess with the grains while they try to settle. Instead. Try to inoculate in different points of the bag and make sure you’re really careful for this part, by injecting towards the screen of the bag so you can keep track of progress
