    @jamalshah881 It depends on which procedure you use. Heating it will help the stain penetrate the cell wall.

    fair play to the lady it simply explains the procedure (not sure why albert is present ? ) yet im not sure about the procedure of using a paper towel which is not subject to crosscontamination, very well explained and visually shown ( cant spell demonstrated)

    so there is no mordant used with the acid fast technique? i thought it was heat.

    No, there is not a mordant. Heat can be used to drive the stain into the mycolic acid wall. It basically depends on which procedure you are using. If you are using the Ziehl-Neelsen procedure you will use heat and steam it for five minutes. I also have a video for that particular procedure.

    Don't you have to use steam for five minutes so that the carbolfuchsin can penetrate through the cell wall?

    @Wressnigg When you heat seal it to the slide, chances are unless you have some endospores or extremethermophiles, you've pretty much killed anything alive on there.
