      lol. right? just buy a bottle of everclear and dilute it with distilled water. ethanol is literally better than iso in about every way.

      @@TM-pi2rn Yeah, I'm using denatured ethanol (branded as fuel alcohol). It seems a little cleaner than most denatured alcohols. They put a sketchy ketone in there though, so I don't put it in a spray bottle like I would IPA. For any spray work, I'm using a 10% bleach solution in water. Been doing this for a few weeks now and no uptick in contamination (knock on wood)

    I love your videos, so helpful. Do you have a recommendation for a water to molasses ratio to propagate spores from a syringe? I think Stamets recommends 5 gallons (80 cups) to 50ml molasses for a slush, which I thought would probably work.

    Hello my friend, your videos are great, I hope you always be successful, I have a question, I use food coloring in agar and this causes the leukoid culture to get a dark color and the mycelium in the liquid cannot be found properly.       Yeah that's a lot for one jar I have problems with to much moisture when I do that

      It's a lot, but it's the fastest grow in I've discovered so far. The jars grow in super fast that way. If you go more than that though you might get slow growth in the bottom. LC is cheap, might as well get speed out of it.

      Btw, started using oats because of you. Now I wonder why God even created other grains! :) But I use a different Tek. I believe it gives me better hydration. Cooking without soaking.

    I just use boiling water from kettle to add to LME to make my LC. I have never cooked them longer than 15 minutes with no contam issues. Maybe its the fact I start with boiling water.

    Have you done a vid where you show how you get from grain spawn into the masters mix? I've seen your machine create the master's mix bags, and the steamer you built, but not the final mixing together. I assume the master's bag is cut open, grain dumped in, and the bag resealed.

    So, after make the grain master, you transfer to growing bag or you made another "grain parents?" Cz here in Indonesia, the grain master we called as F1 then from the grain master, we transfer it to F2 which basically another "grain parents" after F2 we transfer to F3 or we called as growing bag

    Great videos always Andrew! Let’s let one more myth go alright, scalpels are not intended to be flamed red hot. It just needs a couple quick passes. Yeah it may look cool but all it’s doing is damaging the blades sharpness and nothing else. Not a criticism, just your voice is being heard so spread the spores

    I see your pressure cooker in some of your videos, but is that what you use to sterilize all your substrate? You seem to have quite a few bags in your grow rooms. Do you use a bulk sterilzer? If so, what do you use. I haven't had any luck finding much without requesting quotes from commercial suppliers.

      I only use the pressure cooker for culture work and grain spawn. I use an atmospheric steamer for my bulk substrate. You can see a glimpse of it in my fruiting block video.

    Do you have to make sure the blade is cool before cutting into the agar dish? Wouldn't the blade only kill the mycelium immediately next to it, or does putting a hot blade in the dish somehow kill a lot of the mycelium next to it?

      The moisture cools the blade surface off pretty quick, but in most labs we press the blade flat against the edge of the agar to cool the blade before cutting. I don't know if it makes a big difference but that's what they teach people in universities/colleges. You can search microbiology lab culture techniques and find a bunch of really usefull general info about lab tech and procedures right here on youtube and google as well. Hope that helps.

      I just did this. A full plate of agar; 2 ml, 4 ml, 10 ml, and 40 ml liquid culture.
      2 ml was slowest
      4 ml = agar (but nearly the same as 2 ml)
      10 ml fastest (2 days faster)
      40 ml not happy. Too wet and did not fill out the bottom of the jar.

    Hi Andrew, have you ever considered injecting the LC into the substrate. Without the extra step of grainspawn. And if its a nutrient problem one could add rye into the substrate, and sterilize. what do you think about that?

      Yes, I've had mixed results with it. I'm running more trials on it now.

      The grain provides some nutrients, which is helpful if all your feeding the mycelium is carbon.

    Hey I’m just getting started out and want to try and make my own liquid culture. I was curious to know what size syringe and what gauge needle you used to get the tissue before putting it in the jar?

    How do you go about finding what strain of mushroom to grow and where to sell them? Especially now, with restaurants and farmers markets basically being shut down, I am afraid I will be investing money on fungi and have no one to sell to.

    Might i suggest instead of isopropyl alcohol, that you use hospital grade sani-cloth. It's not flammable and kills everything very effectively.
    I would hate to see you get that neat "blue glow" to ya ...

    Hey quick question. I noticed two of my agar plate samples have the slightest blue tint in a spot or two have you ever seen such a thing?Is it contaminated or does it happen because it doesnt seem to spread or anything

      Most likely contaminated. Any mycelium growth

    But the internet says.... that the syringe filters do not provide enough air exchange for grain. I had not read this until after I tried it. The 0.22um x 25mm PTFE syringe filters are working great for me... with both liquid culture and grain.

      Unfortunatelly I had bad experience with syringe filters...I don't know how but when I made like 5 jars 3 with polyfil and 2 with syringe filters I had very very slow growth in my syringe filter ones and when I opened them up it was definetly a carbon dioxid like smell from the jars.
      I don't know why that happened to me. Maybe because these were from china and low quality. Since then I haven't tried.

      There are 4 different types of filter material in the discs. Some melt at pressure cooking temperatures. The PTFE (Teflon) filters survive pressure cooking. Do you know the material in the filters that didn’t work for you?

    MY question is how do you keep it at 15psi for 2.5 hours? You have some type of system at that keeps it at 15? Mine will go up and down and i have to manually adjust

      If u put the oven eye on med it should stay close...u can adjust slightly til it says put. But never leave ur pressure cooker unattended.

      I let my 26Q presto go at full blast until it gets to 15p , then I turn it down to about 3/4, if it stays at 15 after 5 mins ,I lower down to 1/2 flame, if it stays I’ll lower it down to 1/4 . If it goes down below 15, then blast for a few mins then try again. I can usually get it down pretty low surprisingly. It must depend on what ya got in there, jars or bags or lcs/agar.. josh, this is an old video right? U said 2.5 hrs pc? I’ve been pretty darn good with 90 min after it gets upto 15? Nice agar shaking / distribution advice! Never heard anyone talk about that! How to keep it from sticking to the inside of the lid etc… love your tunes , give us yer playlist?!?! 😊😊

    15psi for 2 1/2hrs, so where would the water level have to be inside the cooker to prevent complete evaporation? (I'm new to this stuff, sorry)

      no, it will evaporate, do not soak it, just a light spray.
      using a UVC light is better though

      it's not a ventilator, it's a laminar flowhood

      basically, it moves clean air over your work area and ensures that it has almost no chance of atmospheric contamination

      He does sort of explain it in at least one other video. Maybe the one on LCs . There are 2 pre-filter in the upper column and then a heppa filter where the air flows out. Also would need a good fan in the upper column to push air through.

      nah pressure cooking will sanitize them, pasteurization is a less effective method that people use for straw mostly, or wood chips and such for substrate.. this is for grain spawn, and the pressure cooker does just fine

    Why is it necessary to break up the initial growth of the mycelia? I can see that after breaking it up the grain master is consumed quicker. Do the mycelia reunite before they produce a mushroom? Can you reuse those lids with the injector ports. Once you close the bags is there an air supply to the grain master? I saw on another video there looked like a filter on the side of the grow bags.

      To make it run quicker. Also some varieties will start to fruit in the older section of mycelial growth while the jar is still growing in.

But I keep on thinking if it is anything like growing vegetables you wouldn't want to have too many individual plants in a set area. So do the mycelia rejoin to form a single fungus combining to produce fruit?

    Why not take the edges of the mycelium off that afar dish if the edges are supposed to have the strongest mycelium? That where I’ve been taught to cut from to transplant.

      Yea ive seen people cut out the fastest growing part and put that in a new petridish with agar and repeat it a few times to get the strongest and fastest colonizing mycelium

    You pressure cook those mason jars with that lid setup, but also without tin foil on top?? I've never used those lid or that setup and is why I ask.

      aluminum foil is only really necessary if youre only using micropore tape with punched holes in lids to prevent any water dripping off the inside of the lid of the PC on to the grain jars and into the grain
