    Hey Andrew fun fact. You can use your torch to flash the agar after you pour it. A quick flash will eliminate all bubbles. I used to do antimicrobial textile research and this technique used to help when pouring a few hundred plates at a time. I hope this helps and you see the comment. Best regards

    You should do an epoxy resin pour over that tile table you have there. Looks a bit distracting to the eye because of all the splotches and looks a bit porous. Nice smooth, clean epoxy would look great! Look up epoxy countertop if you don't know what I'm talking about.

    Something cheap and easy to use as a container for making and PCing agar is an empty Pedilyte bottle (or the cheaper Walmart brand). DC Mak taught me this trick, and it really works! The bottles are PP5, and hold up very well to being pressure cooked. Before I had my media bottles this is what I used, and they survived tons of PC cycles without melting or deforming.
    Just a tip for some of you beginners that are on a budget. Theyre like $3 each, and you get a drink out of it too!!

    Hey thanks for this.
    I came here because this video was shared by Alan himself so I thought it must be good. It is good.
    I have two questions:
    How long do you PC your agar?
    What temp do you pour your agar?
    I ask because I have some condensation issues on my plates, specially on the sidewalls and the droplets there then slide onto the agar and slosh around.

    Professor Mossy, Love you and love your video’s. Personally I have had very good results with a plastic pitcher for pressure cooking and pouring agar. But I see the benefit of being able swirl and stir. For centrifuge tube slants, I fill the tubes and then pressure cook. One less potential point for contamination. I can also add a little piece of wood pellet and insure each tube gets the same amount.

      Caps sealed tight. Pull out of pressure cooker after pressure back to atmospheric. Prop at angle till cool/solid. Then I open in front of flow hood, pour out condense, and add a piece of colonized agar. My only contamination was when I dropped a cap and put it back on the tube.

      Don't really need a whole video for that. It's the same exact thing as his other agar videos, just a different agar recipe. 20g of agar agar per 1000 ml of distilled water. That's all.

    1:20 A PTFE lid eflask in a microwave is quick and easy. You can pump out a 1000mL flask (if it fits) in less than 5 minutes.

    I like to pour with a freshly washed long sleeved shirt on no matter what time of year or how hot it is.... idk why.... not really sure if it makes a difference, but I also ended up shaving my head completely bald for this, so I'm just a little overboard. 😆 I was just wondering how to get less bubbles, and now I know, thanks. I got some really good tips from this, thank you.

    I use a 1000ml media bottle with a magnetic stir bar in the bottom and place on my stir plate for about a minute to help blend the agar and lift any sediment from the bottom of the bottle before pouring.

    You just saved me a lot of aggravation.... I will forever use jars of water as a temperature buffer with my glass flask now. I also have a Nalgene flask.... only thing with it is that I can't use the heat on the stir plate with it.... but it sterilizes fine

    So i think a video idea is about pressure cooker maintenance. The seals wear out and sometimes they dont pressure as fast as when their new. This is not for me but for those who are new to pressure cooking. Models that they might wanna look at and buying a back up seal.

    Out of all the cultivators Ive been watching, your mushrooms look the best and you have the best mentality. I fell out of my passion for art after getting my degree for illustration, due to it just not being fun anymore and the whole culture behind the scenes. Ive been wage slaving for almost 10 years thinking I wouldnt find another passion. I grew some mushrooms and just fell in love with everything about it. Im trying to put together a business plan and Ive been trying to figure out just how many pounds I should aim for to start. How big of a space did you start with and how many pounds did you start growing before trying to sell them?

      Just start man, there's a decent learning curve in getting your grain spawn prepped properly, and learning how to setup a grow room for proper air flow and all the little techniques to get certain strains fruiting properly. Just go for it, you gotta start somewhere, just remember you have to think of it as a business, loving what you do is important, but if you put that aspect in front of the business you're most likely going to fail.

    Does the additive (sawdust, etc.) actually have an effect in agar that way? It looks like it has all settled to the bottom by the time the agar sets, which puts it out of reach of mycelium growing on the top surface of the media.

    SUB'd !!!! Thanks for the Video - quick question if you might? getting into the hobby. I've done PF tek with some success. Tried to go giant auto mono-tubs and it just stalls when I go bird seed to bulk substrate with manure, but it doesn't really contam till week 2-3 when I'm poking at it. I think it might have two much FAE because it's a bit dry when I give up, but I would think the healthy seed would at least try to spread. I've increased humidity and still didn't see it trying to colonize the substrate - It just gets fuzzy but doesn't spread. Any tips? I'm going to start over and while getting experience with agar, > going simple mix for bulk and reducing FAE to close to zilch. I have a flow hood like yours and threw a few grand at this so I'm motivated to get it right. lol Thank you - especially if you can find time to respond? lol

    how to you prevent boiled over agar I Kept getting mine boiled over in the flask and it end up with half. BTW your fingers are all over the place ie on the flask lip, inside of the petri dish.

      You want to pour at 117 degrees mate at 107 it begins to gel up quick

    Any particular reason you don't use glass petri dishes? Obviously, you wouldn't use them for the cultures you sell and ship out since they're pricey, but seems like they would be a good long-term investment for internal use. You can reuse them forever and pre-pour before sterilizing, so less risk of contamination.

      @@justinlangley9529 "the added work or cleaning and sterilizing glass dishes is yet another expensive add on."
      There is no "added work". Cleaning is just dumping out the current chunk of agar (you're going to sterilize the whole thing afterwards anyways), and sterilizing pre-poured glass dishes is the same amount of work as sterilizing agar in a separate flask and then pouring into plastic ones.
      "Dont you hear him speak of 'poverty'?"
      Sure, I also hear him speak of constantly having to buy new cases of 500 plastic plates that cost $300 a pop. If anything, buying glass petri dishes would save him tons of money in the long run, which is the main reason I suggested them.
      "Handling glass with gloves x10,000 times a year is another reason"
      Honestly, don't get this one. Is handling glass with gloves worse than handling plastic with gloves or something?
      "Plus the man is a breeder and creator of new commercial strains. In other words...LOTS AND LOTS OF DISHES!!!"
      Which would be another reason to buy lots and lots of glass dishes once and reuse them, instead of buying lots and lots of plastic ones over and over again ad infinitum.

      @@otacon1024 I agree with what you just said, I have all plastic.....and some reusable autoclavable ones still going strong (none broke yet) after 25yrs. They are more costly, but VERY ECO friendly compared to the disposables.

      Honestly man, the biggest reason is because I use pressure cookers and not a sealed autoclave. It wouldn't be a kindess to me to switch to glass right now. One day I'll have the infrastructure to go back to glass, but today is not that day.

      Yes, as far as I know. I've never grown out the black oyster before though.

    Could you fill the Petrie dishes first with unsterilized agar, then sterilize them in the pressure cooker?

      This is an old question, but just in case you're still interested or for future readers, the answer is no. Well, not with petri dishes. You absolutely can do agar work like that, though. It just takes a few modifications to the process first. What you'd want to search for is 'no pour agar teks', and you should find plenty of info.
      It's a great intro to agar work IMO, and helps demonstrate how easy and simple working with it really can be.

    (New to shrooms) ignorant question ahead: What's the purpose of the poly fill? Why is aluminum foil alone not sufficient?

      I'm sure you've figured it out, but polyfil is used for filtration. You can use Synthetic filter disks, double layered tyvek, a few other materials, but it's just to keep any contaminant out of your media.

    Ja tego nie panimaju. Pracuję na agarze w salonie, oczywiście dezynfekcja 70%izopropylem, rekawiczki itp.. Ale zadnego SAB, zadnego floowhooda, i jedyny contamination to 1-2 na 25-30 zrobionych petri dishes.... To duzo czy mało? Bo mi to wcale nie przeszkadza. I zaszczepiając /klonując czy coś jest taki sam waste. Na 25 gotowych, strace 1-2 przez zarodnik plesni. Tyle w temacie xD

    So I know that Agard dishes are disinfected with a gamma radiation but when you’re done with the dish, can’t you sanitize it and sterilize it and use it again

    Wait if you hate yeast why do you used it in your LC mix is the yeast spent by the time it comes out of the Pres cooker?

      The pasteurization kills it off, i beleave he said in a previous video, its cause he sees it as a good supplement for his fungi, like a multi vitamin.
      It may also have to do with preventing enzyme blindness(like adding some masters mix), and promoting his fungi to express/strengthen enzymes that would allow it to fight contamination when in production.

      They're sterilized pre packaging, irradiated at the facility that makes them, that's why he had to tear it open before he started.

    I did a test with agar plates using dye vs no dye and all my cultures grew better without the dye. Only thing different was the dye. I will never dye my agar again.

    i didn't know you could use liquid nitrogen to store mushrooms. You might could find save some money by buying some liquid nitrogen tanks from select sires or other livestock organizations. Our breeding guy has a liquid nitrogen tank to store semen we've bought and then to breed heifers. You could store a lot of strains of mushrooms in a tank like that.

      It's recognized to be pronounced either way. Either with long A-gar, or as AH-gar.
