    Antipsychotics, also known as neuroleptics, are a class of drugs used to treat psychiatric disorders, such as schizophrenia, bipolar disorder, and psychotic depression. Antipsychotics work by changing the way certain neurotransmitters (mainly dopamine) act in the brain. This pharmacology lecture covers topics such as schizophrenia, role of dopamine in the development of psychosis, dopamine pathways in the brain (Mesolimbic, Mesocortical, Nigrostriatal, Tuberoinfundibular), positive & negative symptoms of schizophrenia, dopamine & serotonin receptors, mechanism of action and side effects of antipsychotic drugs (1st generation typical & 2nd generation atypical). Drugs mentioned include; Haloperidol, Fluphenazine, Prochlorperazine, Trifluoperazine, Chlorpromazine, Aripiprazole, Clozapine, Lurasidone, Olanzapine, Quetiapine, Risperidone, and Ziprasidone.

    Not a student, just trying to educate myself on antipsychotics after being prescribed for a few years, I had psychosis. Currently weaning off of olanzapine.

      I'm on risperidone after psychosis in 2018 and 2019. I lowered my dose from 4 to 1 mg over the past 1.5 years. How's it going with you so far weaning off the olanzapine?

      Awesome that you are weaning off, I currently have minor form of PTSD, self diagnosed, and have been prescribed olanzapine, methex od etc.

    my life was for a long time ruined by antipsychotics. was prescribed them and although I suffer from major depressive disorder, I do not have schizophrenia, bipolar, or any sort of mood disorder. Because I already lack serotonin and dopamime due to my depression, this medication sedated me/turned me into a zombie, made me gain 15 lbs in a matter of 2 weeks, stripped me of my personality, confidence and creativity, & even after I went off of them, I could not get back to the girl I was before. I no longer was responding to any antidepressants that had worked in earlier years or stimulant medication that had helped in the past. I looked awful and became a different person. All I wanted to do was sleep. I was so severely depressed i could not function at work (yes, I was trying every class of antidepressants at this point). I was no longer myself and had become completely overcome by my mental illness. It got so bad that my therapist insisted I seek more comprehensive medical treatment. I admitted myself to the hospital and after many more trials of medications, I had to resort to electroconvulsive therapy. I am 23 years old. Although it has a terrible rep, this treatment saved my life. the reason I am writing this here is because I think antipsychotic medications are necessary for people who naturally go into manic states, suffer from schizophrenia or psychosis. however, I am writing this as an informative warning that these medications WILL change you, and WILL sedate you. for some this is necessary. however, if you are prescribed these because antidepressants alone are failing, I really hope you will do the research on how they work because I believe that for a depressed person, these meds can be extremely detrimental . I hope that those who need these meds find them helpful. However, for me the damage they did could only be repaired by literally shocking my brain out of it. Good luck to all and please know YOU ARE NOT ALONE.

    I was misdiagnosed with schizophrenia. Been drug free for a long time and I am still suffering the disabilities caused by these medications. I cant express emotion and have slurred speach. The eay I walk changed. My hormones are all over the place. I exercise and hope for a better future. I feel like my a bird shose wings been clipped.

    Most people will never experience extrapyramidal symptoms (EPS) so they have no idea how horrible they can be. There are many factors contributing to EPS but mainly a dysfunctional dopaminergic and acetylcholinergic neurological system play a role.
    There are so many hyperkinetic movement disorders like dystonia, dyskinesia, akinesia, akathisia, athetosis, ataxia, chorea, myoclonus, ballismus, tics, tremors, Tourette’s, spasms, RLS,etc.. it can make it very hard to distinguish.
    I sympathize for schizophrenics and people who need certain antipsychotics and other medications that exacerbate these movement disorders. Most these medications drastically deplete Dopamine which is one of the main reasons for these extrapyramidal side effects.
    Science has had more than enough time AND MZoNEY to develop a novel medication that should not cause this. Many schizophrenics need glutamate and dopamine regulation (not always “reduction”). There are several dopaminergic receptors and binding sites that can be antagonized and agonized therefore there is ZERO reason why there isn’t a proper neuroleptic to control psychiatric side effects while not exacerbating extrapyramidal side effects OR anhedonia!
    Dopamine agonists can do wonders for EPS and science was able to develop non-ergot agonists which are much much safer yet can’t develop an agonist that will not cause debilitating Dopamine withdrawl Syndrome? Come on that is ridiculousness there is no excuse for no development of one. Or how about an MAOI with no withdrawal? Nope, doesn’t exist 🤦‍♂️
    9-Methyl-β-carboline should be further examined as it appears to have incredible dopaminergic restoration properties. A study was done on rats that proved it. In layman’s terms scientists injected rats with synthetic neurotoxic compounds like 2,4,5-trihydroxyphenethylamine (6-OHDA) and 1-methyl-4-phenyl-pyridinium ion (MPP+) that selectively destroy tyrosine hydroxylase and dopaminergic neurons. So they essentially gave these rats Parkinson’s like symptoms.
    After a few weeks of 9-Methyl-β-carboline administration around 80% of the damaged neurons returned to normal determined by measurements of nicotinamide adenine dinucleotide dehydrogenase activity.
    Also the adamantane derivative called Bromantane and Pentadecapeptide BPC-157 play important roles.
    Yet no ongoing research is being done. 🤦‍♂️

    thanks for this but just to note a MAJOR correction: psychosis is associated with increased dopamine presynaptic function in the nigrostriatal pathway (particularly the part that projects to the associative subdivision of the striatum) it is NOT the mesolimbic system

    Remember 'Antipsychotics' can be 'hyperbolic' - which means that withdrawal needs to be approached more carefully at reduced doses.
    And remember that abrupt 'antipsychotic' drug withdrawal can result in 'Withdrawal Psychosis'.

    I think the APs causing onset or worsening of negative symptoms is called “Neuroleptic-induced deficit syndrome”. I feel like this is something that needs to be talked more about!
