  • APOE2 is an important protein in the brain. APOE2 has been found to increase longevity in people as well as animals. APOE2 has been found to reduce the risk of Alzheimer's disease. The video is a requested video about doing a deep dive into APOE2. Overall, APOE2 reduces total cholesterol and increases activity which could be why people with APOE2 live longer on average.

    Now the key question would be about the interaction with nutrition: caloric restriction, low carbs crossed by high monounsat fats. Would E4 mice live longer, would E2 mice lower their triglycs ???

    My dad is an ApoE2/E4 and my mum is an ApoE3/E3.
    My two sisters are both ApoE2/E3, but I am an ApoE3/E4 🙁
    The diet for ApoE4 is the hardest for me to follow.
    I do not mind eating raw oily fish that they serve in Japanese restaurants, but they are costly and they do have mercury and PCBs if you eat it too frequently.
    I tried to eat olive oil salads with avocados and lean chicken breast without saturated fat or carbohydrates for a while but I failed because it was not palatable.
    The lowest I ever got my cholesterol and LDL-C to purely by diet alone were 168 mg/dL and 86 mg/dL respectively, and I fell off of that diet after a month and everything went back up.
    I am considering taking medications to keep my cholesterol low so I do not have to be so restrictive on what I eat.

    Wish someone would do a video on people who have Apoe 2/4, like me. I struggle with cholesterol, triglycerides, etc even when eating a really healthy diet. I’m 46. Just from the couple published studies I’ve read thus far, the 4 overshadows the 2. And I’m not sure the 2 even helps you if you have the 4 also. I have constant brain fog, headaches, etc. One published study I read from Alzheimer’s website said 2 is not totally benign and has increased risk of neurological disorders and cerebrovascular disease.

    What dietary factors effect triglycerides most significantly? The graph showing the elevated triglycerides for the 2 variant has me thinking.

    What is the deal with ApoE4/4 on the Kaplan-Meier graph in the review paper? The rates of Alzheimer's are much lower in 4/4 compared to the 2/4, 3/4, and 3/3 combinations (2B). However the OR graphs tell a different story with e4/4 toping all others by a large margin (2A).

      That graph is not right. In the actual paper, the colors are different. 2/4 doesn't show up on this one at all. The black one is 3/3 and the bottom is 4/4. I'm not sure what happened here but you can check the paper. It's called "Exceptionally low likelihood of Alzheimer’s dementia in APOE2 homozygotes from a 5,000-person neuropathological study" Fig. 1.

    In detailed subgroup analyses, APOE ε2/ε2 and APOE ε2/ε4 genotypes were most consistently and strongly associated with cortical superficial siderosis (cSS) presence and severity, a central MRI biomarker in cerebral amyloid angiopathy (CAA).

      Yes, nothing is a complete superpower with some drawbacks. I think the paper said only about 5% of people with those genotypes are likely to get CAA.

    Is there a possibility that the macroglia "shift" from making one form of APOE to another in a epigenetic manner? If not is it possible to use CrisperCas9 to modify the chromosome to produce APOPE ε2 instead of the less useful APOE ε4 ?

    Thank You again. I guess I have to wait a few decades to find out, who will win: My "bad" APO3 and APO4 alleles or my SIRT1 and FOXO3 ones, where SIRT1 is said at least to decrease the Alzheimer's risk while APO3/4 increase it. Meanwhile I must check on my HDL and TG-Levels. Hope to understand immunology still in 20 years ;-)

    Interesting you posted this because just the other day I was looking at how keto diets effect microglia. It's way over my head this stuff but it seemed like there is some connection between apoe4 and that a keto diet is beneficial for AD patients with A4. However it might be destructive if they don't have A4 or are only A4 gene carriers.

      I heard the E2 will offset the E4 effectively making you a E3/E3. I believe it was Dr Perlmutter.

      Unfortunately not. So much of the issues with prion proteins happens inside of cells where antibodies are no use.
