  • A suggested method to cover microscope slides with paper for longer durability and value. Paper-wrapped microscope slides were common in the 19th centruy. Here I try to rediscover how it is done.

    He is so excited every single time. I love the smile on his face that appears on every single video. He doesn't seem like he is doing this because he has to, he is clearly having fun in the process

    Wow! I learnt something new! Despite having used microscopes for many years (school, college, professional college, and pathology years, I had never known of paper-wrapped slides! Thank you professor for teaching me something new! Great information indeed!! You have really immersed and shared with others your learnings and experiences! Bravo!! Thanks, once more!

    After fooling around with the white glue I tried spray contact cement. Hold the can about a foot up and put a thin layer onto the paper. Let it get tacky and it works great, instant hold. (be sure to be accurate with the placement and PROPER SIDE UP!) Also, you can easily wipe off the slight overspray of the lacquer on the window with a tissue soaked in acetone. If you use the yellow tack note paper for making the cover paper when you do the spray you don't get any leakage and it peels right up easily.

    I recommend something called “ART GLITTER GLUE” and be sure to purchase the fine tip that they sell alongside the glue. It is not glitter glue. The brand is Art Glitter Glue (very popular for scrapbooking) The fine tip is about as small as that of a mechanical pencil which allows you to put the smallest amount of glue needed (perfect for the edges that get folded over). And if you are only cutting paper with the hole punch it would be much more cost effective to just purchase a paper punch at any crafting store (Hobby Lobby, Joan’s, Michaels etc.)

    You might want to try using drafting dots to cover the window. They’re made out of masking tape, and should cover it well without allowing any paint underneath.

    This is nice. You could use liquid tea to stain white paper to make the paper look aged to give it an 18th century look about it.

    I would be hesitant to punch over ceramic tile even with a few sheets of newspaper to protect it. Using some plywood for additional protection would prevent cracking a tile

    Perhaps you could find a spray adhesive that could apply a thin even coat with less excess mess than white glue.

    Amateur microscope enthusiast here. I've been making slides for my 5 year old, catching bugs, and making slides only using book tape and home made card stock slide (rectangular piece with a hole in the middle, taped on both sides and bug sandwiched in the middle). How long would my slides last?

    • @Microbehunter
      @Microbehunter  2 ปีที่แล้ว

      Make sure that the insect is completely dry before mounting (or allow moisture to escape after mounting). Then it should last a long time. Dry insects in display boxes also last a long time.

    Does acid free paper need to be a consideration? Would contaminants in the paper migrate under the cover glass and damage the specimen? What glue is used to hold down the cover glass?

    • @Microbehunter
      @Microbehunter  5 ปีที่แล้ว

      Maybe for long-term storage, acid paper might be better, but here we talk about a very long time. Contaminants will not migrae under the paper: the paper is dry (they therefore can not migrate), beneath the cover glass there is a mounting medium. If stored dry. In humid environments fungus might form, just like with any paper. Problems might be there, however, if the mounting medium is glycerine jelly (not fully solid) and if the cover glass is not sealed (ringed). But then this is a general problem, and not one of the paper. Finally: contaminants are present anyway, with or without paper. I used white wood glue for the paper, but others might also work.

    Have you tried using double sided mounting film for sticking the paper to the glass? I have used this for mounting photos and graphics to display boards. The material I used comes in 648mm x 25m rolls which would be rather more than you need, though it is a useful product to have around. www.lionpic.co.uk/p/32357/SeaLion-C6-Twin-Release-Cold-Mounting-Film-648mm-x-25m-roll
    [edit] these double sided A4 adhesive sheets from EBay may be more practical:

    What I mean is can I use a slide like this and fill the circle with sample of known quantity in order to.be able to count cells?

    • @Microbehunter
      @Microbehunter  3 ปีที่แล้ว +1

      The liquid would soak into the paper. There could be ways like using separate cover glasses of defined thickness as a spacer, or using a recision pipette to take up a specified sample of liquid with cells.There are certainly improvised ways. But for routine use I would rather spend a bit on a hemocytometer slide because they are properly calibrated and the results obtained can then be also shared and published. You might get DIY solutions also to work, but the question is if it is worth it. Eg. take a defined volume and count all of the cells under the cover glass etc. Glue two cover glasses (0.17mm height) with nail polish on slide and then place 3rd cover glass like a bridge across, for a defined volume etc.

    • @irishguy200007
      @irishguy200007 3 ปีที่แล้ว

      @@Microbehunter does a slide exist with a recess that can take a defined amount of sample. I know that the automated cell counters have their own slides that take usually 20 micro liters of stained sample but any time I look up microscope slides I get hemocytometers which have etchings in them for manual counting under a microscope. Could I use a hemocytometer for automated counting using a microscope and phone software or would the etchings interfere or mess up the image captured by the software?
