    Hi Man! Thanks for this very usefull vídeo (as all others!). I guess I had asked you or some other grower just this same question. And now You bring us a very detailed answer! I watched an older video in wich You show us the clamp connections and afther that I decided to by a second hand mycroscope and got to see the God dam clamps by myself! It's really a time consuming economy to have one.
    I've watched a lot of guys on YTB over the past 2 years and I sincerelly say that You are one of the tops. Few growers give such precise information. I have two questions, hope You can answer please.
    - Do You have some notice of people growing PCs on coffee ground?
    -Does mycellium contains psilocibine (I've reard some comments that say yes)? If yes, why it don't turns blue? And more: why whe don't consume the mycellium instead of waiting long weeks for fruit harvesting?
    I am asking this because I am tryng to grow a single fruit for more than one year and didn't reach the goal...yeat. But at least I've learned (including watching Your channel) almost all the prior steps to get their: agar, transfers, asseptic conditions etc. I hope I will get there soon. Thanks!       Glad you enjoy the content. I'm also glad you got a microscope. You never know when a child or some family member will want to use it. Something that will find uses throughout life. Not only for our babies.

      >>I've never hear of using coffee grounds for cubes (I'm assuming that is what PC is :) ) I have used them to grow oysters and personally think it is a waste of time. People were feeling guilty about their coffee grounds for years, but in reality they don't help and you can just compost them.

      >>yes, it does. You can extract it and use it. I have personal proof and so do many of my friends. If an HPLC doesn't detect it, that is a problem with the extraction of samples or the machine.

      >>Mycelium does turn blue as it ages. Young mycelium might not have matured enough to produce the alkaloids yet.

      >>you can consume the mycelium. Look up my "Purple Drizank" video. I have extracted and used extracted mycelium many a Friday nights. It is a good use for old or excess spawn and 100% has an effect. Use any acidic juice and steep it just like you would tea. Put some marbles in it, and give it some shakes during the process, store in the fridge, and consume within a few days. It WILL oxidize!
      Keep trying. You will eventually succeed. Mycology is a life-long hobby for me. There are going to always be setbacks like everything in life. Cheers!

      >> I know a lot of people were using coffee grounds to supplement their substrate in the past, but I don't think if made much difference. You can try, but I'd keep it under 10% of the total substrate.

      >> Yes it does. I have a video about 'Azure drizank' that tells you how to make a drink using old spawn. The alkaloids will be much more concentrated in the fruit because they are essentially big wads of mycelium matted together.

      >> It turns/bruises blue because of the same chemical reactions that make the fruit bruise blue. It kind of complicated, but it's a type of oxidation and polymerization reaction. There is a good paper. You can search for "Injury‐Triggered Blueing Reactions of Psilocybe “Magic..."

      >> The fruit are just an easier (and nicer) way to consume it. Also more concentrated.

    hey bud, i appreciate your expertise and was wondering if you could give me a recommendation(s) for a good scope, what manufacturers to look for and what magnification to get (at minimum) for mycology purposes - obviously. I thank you in advance for your time! --- nvm, i wrote that before i saw the part where you mention the amsope, that the brand to go with?

      I've had an Olympus for CX21 for years. If I had to buy a new one, it would depend on my budget. For $250 ish (no camera), I'd go with the Amscope B120C. For about 4X that, you can get an Olympus CX23. CX43....well... Depends on your budget.

      @@edwardgrand i make questionable decisions - as a result, my budget varies and is often ignored. I do like the idea of having a camera. You can use it to take snapshots and then zoom in and examine those correct? or does it just take a photo of what you are seeing? (i guess a better question, does it have high enough resolution so you can zoom in effectively)

      @@etownKeystoned The 100X objective (x10X eyepiece) on the Amscope B120C is far more than enough magnification/resolution to see any microscopic character you would need to in Mycology. I don't find cameras on scopes necessary for anything. If you want to snap photos or do videos, you can buy cheap $5 phone mounts online.

    Hi Ed..for my purposes I’m really only chasing dikaryons to expand further. Do you think mashing some samples of the swab together in one area may be better than the grab ‘n’ drag? Thanks

      I also like the 'grab 'n drag' for getting dikaryons too. If you do it, but just let them grow together, you can pick something that is going rhizo already. I also use the T0 plate to make multispore spawn and do some phenohunting when they fruit. The cool thing about any streaking technique is it allows you to separate the good spores from any contamination that might be on the swab. If you just mash the spores together, and there is any contam in there, it will take over before you can isolate a dikaryon.

      Nothing wrong with ghetto swabbing. I am sending monos in cryovials now. I can send some to you if you'd like. It's a lot more intense than just having the monos. The real work starts after you put two of them on a plate. Ghetto swabbing is full of mystery, but is a lot more straightforward I think.

      A healthy monoculture should grow concentrically. It is the nature of mycelium to grow out from a central inoculation point. Like fairy rings you might have seen in the grass.

      Could potentially come across a dikaryon that grows on one side/not concentrically? why would that happen? would it be better to toss it and find a more concentric candidate?

      @dani26191 it really depends on several things. But generally, isolated, filamentous fungi grow that way.

      No international issues. I ship all over the world. Never have any problems. For 12 sets, I'll throw in 4-5 extras. Hard to do codes with the website. Logistics issues.
