Hi, Ed! Do you have a preference between colored or non-colored agar for microscopic viewing of mycelia? specifically when checking for clamps? Thank you for everything you do!
No preference. The key to getting a good image is to close the condenser/aperture to get a good depth of field. You will have to increase the light intensity. Stopping down the condenser/diaphragm also gives the clamp a different look because of the diffraction. I don't think the color has much to do with the microscope view. It just makes the plates look pretty and it's easier to see colonies.
Thanks for this microscopic view and talk. It helps get a little bit of understanding of mono/di karyons than talking or reading alone.
with such a small sample being viewed, and the low number of clamp connections found in this sample. What would be the next step to get the dikaryons to spawn? the sample taken is destroyed, what if the sample of that same plate I put to grain have an absence of clamps or contamination... While this video helped a lot with what to look for with my new microscope, I'm still a bit confused on best practices as to what agar to take to spawn .
I subculture very small pieces of agar from the leading edge of colonies. By T3, it should be a monoculture.

I use an older Oympus. I'm looking at the Amscopes now. I think they're a much better deal. Optics are good enough for what we do and most anything to be honest. $250 I think is a good place to start.

I love this! I have a 1000x microscope I bought for fun years ago, but I never had anything “productive” to use it for. Just to satiate my curiosity. It’s been tucked away for years now, and I just got into growing mushrooms, so it’s perfect 😁
You can start looking at stuff from the wild too. Some spores look really cool under 1000X.

Maybe not "exciting", but incredibly valuable to hear your thinking as you scan this slide. For people like me w/out a biology background and who have never struggled with using a microscope for real work, understanding the practical context and difficulty of this work is crazy helpful. I might be alone here, but I'd love a session where you walk through the basics, along with some of the uses of a scope in a mycology lab. What zoom x is this video done at? What's the ideal to look for clamps and look at spores?
Most of that was at 400X, but I think I had to zoom in with the camera phone to get a decent shot.
Clamps are viewed at 400X. Spores should be viewed at 400X if you are just checking for concentration or presence/absence. 1000X for spores if you are looking for ornamentation or measuring them with an ocular micrometer.
