I have a 2020 Sienna with a manual rear door. I'm puzzled same as you. Maybe the whole interior trim panel has to be taken off?

​​@@williammiller402I'm sure you probably figured it out...but if you manually push the door shut..to where it's hitting the metal latch on the hook and not closing, then push the door open button above the license plate (like you are opening the door from the outside) it will open the latch up so it can properly close.
I had my car on while I did this just an FYI.

yup you can reach inside and wiggle the door open . There seems to be two moveable levers in mine and it is the lower one that opens the door.
