    thank you for the video, why do you decide to resuspent the cell pellet in 10ml media? will it affecting the calculation if i use different amount of media to suspensing the cell pellet?

      I also have this question. Because in my lab I usually resuspend in 1mL, if I want to plate in larger plates and not like the 96 well plate. what determines the volume of medium to resuspend the cells after centrifugation. Also I like counting the cells just before centrifugation. Does this affect anything?

    is there any simple process to know the confluency of the cells? i always get wrong in judging confluency in my lab? we donot count cells for subseeding, we just watch the confluency to be between 70-80% and then subseed it to 20%, is it right process?

      Different labs have different protocols, so there are various methods. I think the reason for determining the appropriate number of cells per well and counting cells is to ensure consistency when comparing the differences in efficacy of substance treatments.

    계산식을 보다가 이해가 가지 않아서요
    1. 9.8x10^5cells/ml:10ML=1x10^4cells/100ul:xML
    따라서 xML=((1x10^4cells/100ul)/ (9.8x10^5cells/ml))x10ml이 아닌가요?
    어디부분에서 잘못한건지 답은 틀리는데 어떻게 이해해야할지 모르겠습니다.
    비례식을 썼는데 10ML 이 어디로 간건지.. 헷갈립니다.
    그리고 뒤에 6well seeding 과정 설명에 100well 필요한 cell 이라고 적어주셨는데 7well 기준 필요한 cell 양을 잘못 적으신건지 궁금합니다.

      답변이 늦어서 죄송합니다. 영상을 검토해보았는데 제가 잘못 표기한 것 같습니다.
      Cell suspension 10 mL 중 Live cell 1. 9.8x10^5 cells/mL 이라서 표기가 (9.8x10^5 cells/mL) x 10 이 맞습니다.
      비례식을 정정하자면,
      (9.8x10^5 cells) x 10 : 10 mL = 100 (필요한 well 수)x (1x10^4 cells) (well 당 필요한 cell 수) : X mL (Cell suspension 10 mL 중 필요한 cell 양)
      10 x 100 x 10^4 = (9.8x10^5) x 10 x X mL
      X mL = 10 x 100 x 10^4 / (9.8x10^5) x 10 = 1.02040816 mL
      입니다 ! 혼란은 드려서 죄송합니다 ㅠㅠ

      평소에 seeding을 할 때에는 아래와 같이 비례식을 축약해서 사용하는 게 편할거예요.
      Cell suspension 중 필요한 cell 양 (mL) = (well 당 cell 수) x (seeding 할 well 수) / (Cell suspension 중 Live cell 수)
      X mL = (1 x 10^4 cells) x 100 wells / 9.8x10^5 cells = 1.02040816 mL

    선생님께서는 seeding용 plate와 culture용 plate를 따로 만들어서 실험하시나요? 아니면 seeding하고 남은 cell을 culture하시나요?

      Culture용 dish (maintain dish) 는 고정으로 계속 Culture 를 하고 seeding이 필요한 경우에는 subculture 하고 남은 cell 을 이용하여 seeding을 하고 있습니다 !
