    Hii I am having a doubt regarding, as live cells= 980000 this value is for 1 ml but you have sample volume 10 ml so total no. Of cells must be 980000*10= 9800000 in 10 ml. Or 9.8*10^5 is the total cell count for 10 ml cell suspension?

    안녕하세요! 댓글 읽어보니 파우더 형태의 추출물을 얻을 때 동결건조 하신다고 하셨는데 어떤 용매를 사용하시는지는 모겠지만 유기용매도 사용 가능한 동결건조기를 사용하시는 건가요?

      @@김정민-j9h 본 영상에서는 추출 시, 에탄올을 사용했습니다. 동결건조기가 액체를 승화시키는 원리라는 건 알고 있긴 한데 적용 가능한 액체가 나눠져 있는 지는 잘 모르겠습니다 ..! 회전증발농축을 하면 농축 과정에서 대부분 유기용매가 먼저 날아가기(증발하기) 때문에 남은 소량은 액체는 수용성에 가깝다고 생각을 합니다 !

      방금 검색해보니까 동결건조기 사양에 따라 컨트롤 할 수 있는 용매가 다른 것 같습니다 (용매의 어는 점이 각각 다르기 때문에)

    안녕하세요! C2c12세포를 이제 막 사용하게 되서 여러 컬쳐방법 찾아보다가 질문남깁니다!! 혹시 100파이 디쉬에 몇프로 찼을때 서브컬쳐를 하면 되나요? 셀끼리 붙으면 분화가 서서히 진행된다 해서요!!

      @@뮤뮤-l3o C2C12 처럼 분화하는 세포의 경우, 세포 컨디션에 따라 분화 상태가 좌우되기 때문에 평소에 잘 관리가 필요한 것 같더라구요 ! 저는 평소에 100 pi 기준으로 70~80% 찼을 때 subculture 해주고 있습니다 !

      @@뮤뮤-l3o 분화가 되려면 거의 세포가 100% 이상 confluence 여야 해요 ! 단순히 maintain 용도라면 70~80% 에 subculture 해주셔도 괜찮을 것 같아요 ! 오히려 세포가 너무 적으면 잘 안자라더라구요.

    안녕하세요 계산식을 보다가 이해가 가지 않아서요 1. 9.8x10^5cells/ml:10ML=1x10^4cells/100ul:xML 따라서 xML=((1x10^4cells/100ul)/ (9.8x10^5cells/ml))x10ml이 아닌가요? 어디부분에서 잘못한건지 답은 틀리는데 어떻게 이해해야할지 모르겠습니다. 비례식을 썼는데 10ML 이 어디로 간건지.. 헷갈립니다. 그리고 뒤에 6well seeding 과정 설명에 100well 필요한 cell 이라고 적어주셨는데 7well 기준 필요한 cell 양을 잘못 적으신건지 궁금합니다.

      답변이 늦어서 죄송합니다. 영상을 검토해보았는데 제가 잘못 표기한 것 같습니다. Cell suspension 10 mL 중 Live cell 1. 9.8x10^5 cells/mL 이라서 표기가 (9.8x10^5 cells/mL) x 10 이 맞습니다. 비례식을 정정하자면, (9.8x10^5 cells) x 10 : 10 mL = 100 (필요한 well 수)x (1x10^4 cells) (well 당 필요한 cell 수) : X mL (Cell suspension 10 mL 중 필요한 cell 양) 10 x 100 x 10^4 = (9.8x10^5) x 10 x X mL X mL = 10 x 100 x 10^4 / (9.8x10^5) x 10 = 1.02040816 mL 입니다 ! 혼란은 드려서 죄송합니다 ㅠㅠ

      평소에 seeding을 할 때에는 아래와 같이 비례식을 축약해서 사용하는 게 편할거예요. Cell suspension 중 필요한 cell 양 (mL) = (well 당 cell 수) x (seeding 할 well 수) / (Cell suspension 중 Live cell 수) X mL = (1 x 10^4 cells) x 100 wells / 9.8x10^5 cells = 1.02040816 mL

    thank you for the video, why do you decide to resuspent the cell pellet in 10ml media? will it affecting the calculation if i use different amount of media to suspensing the cell pellet?

      I also have this question. Because in my lab I usually resuspend in 1mL, if I want to plate in larger plates and not like the 96 well plate. what determines the volume of medium to resuspend the cells after centrifugation. Also I like counting the cells just before centrifugation. Does this affect anything?

    Hi, i'm from Brazil ! I'm having some difficulties with the transfer! How many millimeters is your gel? Here for 1.0M gel, semi-dry transfer was carried out at 24v for 15 minutes. I tried this for 1.5M gel, but it didn't transfer at all. I saw that you use 25v for 1 hour, what percentage and millimeter of gel is that for? I tried to see in the video, I think your gel was 1.5M, but I'm not sure... I also have to transfer high proteins like 250 kDa, i don't know if 25v por 1 hour helps... If you can help me... I would be very happy. Thanks

    how to detect protein marker i am just getting band only no protein marker till 10 min exposure?

      Did you check the detection condition of chemiDoc ?

      @@hyejulee7072 i ran using chemidoc xrs system using chemiluminescense option, here i can only see band of my protein but the marker is absent

    I add TRIzol to cells in 24- well plate to extract protein and RNA and I have 4 repeat for each treatment, but unfortunately the amount of RNA extracted from each well is very different from next well. Do you have any idea what I could do to resolve that issue?

      The amount of RNA may vary depending on the number of cells seeding the cell. Next time, How about seeding on a 12well plate rather than a 24well plate?
