Since people are sharing... I built a laminar flow hood for the cost of the filter, $100. Repurposed the blower motor from an old downdraft kitchen island cooktop, and some discarded white vinyl wrapped kitchen cabinet doors. I'm not a braggart but i'm proud of that. I lust for an autoclave. So tired of pressure cookers. Have some idea to make a pressure sensor to control a hotplate with a timer, so i can set it and forget it. Yeah, it's not a good idea. Also I set my first cardboard box SAB on fire with an alcohol lamp. Fortunately, no one saw me throw the little inferno onto the suburban front lawn.
It's a good idea just put a pressure release valve for added safety. But tbh I got mine dialed in most of the time where I just set the knob and forget it. I usually run it hot af until it builds up pressure and then immediately turn it down to a set level that I eventually discovered works with my burner and cooker and just walk away and I can tell if it's burning too hot by how much the weight wobbles. You don't want it going crazy and a small wobble is all you need. The main thing is you need to run it hot enough to get the pressure built up because it won't reach that pressure at the pressure maintenance heat level. Once you do it enough times it becomes second nature.
I totally relate! I caught my homemade Still Air Box on fire too…although I was using a mini torch to sterilize tools and I would hose down the inside of the box in alcohol between uses. Things were more prohibitively expensive back then so I did what I could but not using an open flame is honestly for the best, if at all possible.

As a mushroom grower. We basically have the same setup. If you need cheap lamilar flow hood look for mushrooms growers. We usually are forced to build it ourself so we are pros at it xD

Aliexpress is a good source of very cheap equipment and consumables- quality can vary but typically cheap enough that the occasional bit of rubbish isn't a big deal
When I was a teen I took some of my moms houseplants and took cuttings. I put them in little medicine cups with different nutrients in them and the one with the most root growth was the one with artificial sweetener. Don't remember what kind. about 1980. This is actually what i did when I had the house alone for an entire week. Party.

using a thermometer or just setting the configurations right turns out is very easy to configure a induction heater for Just the right temperature .... and they are REALLYA cheap, like daam china the forced labour is off the chart
Auctions are great if you are handy with repairing things. Many of the items sold there have one or more issues. Since auction purchases are typically "as is, where is," there is no returning something that's broken. I have a lab instrument service and repair company, and we service a lot of instruments that were purchased by uninformed customers. I would also recommend, before placing a bid, call the manufacturer and make sure they still support the model you are considering.
Came across this vid I use to do this as a hobby like a decade ago. Very time consuming to see the effect but I did really enjoy it. Was in my early 20s limited budged and big dreams. My education or job never had anything to do with anything like it so it was very hard for me at first. Did build my own "clean box" out of a old fish tank to do everything inside and sterilizing it with alcohol before. All the gel media made on a normal gas stove with a normal pot so the steam, PPE and PPM (Plant Preservative Mixture) was my way. Did build a little primitive but working test tube shaker. And use to sterilize the tweezers by dipping them at first in IPA and setting them on fire. Great times. May come back to it agin.

Hard drives go spin very fast. Fans also spin fairly fast. If you want spinny lab devices of either variety, I recommend these. Be safe with hard drive spinny! If it breaks the shrapnel may go fast enough to punch a hole in you.
I'm not into biology, but it has some resemblance of electronics labs, i turned into electronics land :) You have the same desire to do your thing as people in my field have, good to have you on the same planet :) One can see the happiness.

This is great, I am about to build my lab again, but it's specific task is for mycology bulk inoculating. I am going to try and make it as small as I can so it can be on a porch, probably 6x6ft with a pass through atmospheric steam sterilizer in the wall so I can load large batches of substrate sterilize and open it up clean inside. It is made out of a Aluming baking proofing rack with 24" aluminum flashing and foodgrade high temp silicone. The bottom has a steam pan with a 2000watt water heating element and stainless steel float valve. I used to do spore to grow, but the amount of space for dishes, slants and fridges is to much upkeep and space. I dont miss dealing with flaming rubbing alcohol which is why I've outsourced that aspect of the process, just make sure you have a small can of fire extinguisher, they make aerosol sized. This video is defiantly getting me excited to start lab work again. It's a good skill to know for niche product productions, but someone like me who has moved twice this year, it's hard to have space for a lab, particularly because houses in general are almost worse for spore load than outdoor air. I used to do my mycology outside at 2am when the air is still back when I started, because that is the calmest time with the lowest spore load. That can be dealt with by having a well made room and positive pressure HEPA filters though.
Thanks for posting this video. I have a Corning stirrer/hotplate that costs about the same as the Fisher isotemp you have. I think the reason for the high cost of these models is that, at that price point, they come with a temperature probe so you can have it monitor and adjust the heat output to maintain a specific temperature. It's not a feature I've used a lot, but it has come in handy at times. The other thing I can comment on is the Ohaus Scout vs the cheaper scale that you mentioned. I have both and I much prefer the Scout despite the higher cost. The really nice feature of the Scout is I can leave it running all day and it doesn't drift like the cheaper models tend to do. Mine came with a power supply, so I don't have to worry about batteries either.
For sterilizing metal tools such as forceps and inoculation loops you can just heat them to a visible glow in a gas burner, I find this to be easier and safer than the alcohol lamp sterilization

Well done. My efforts don't rise to the level of your efforts; I use a selection of test tubes and glasses of warm water with corks for the test tubes followed by decanting. I fool with sents and flavorings. I have been thinking of trying for distillation, though. Thanks for your advice and insights.

Love your channel!!! I have multiple 23 quart PCs and two 53 liter industrial PCs nowadays, but I started with an insta pot several years ago. They usually achieve 10psi and they work just fine. The time must be extended to get the same results IE a ratio of 1:1.7 is the technical adjustment for the loss in pressure
i have more respect for a video gamer cause at least they arent torturing plants like this. we shouldnt alter plants when we dont know plant physiology. they are living and sentient organisms.

Watching your video I mentioned, that some parts you use could be even less expansive if they will be 3d printed (however it will take some time and effort). Anyway, good luck in all your projects!
My hobby is electronics. I plan to have a laboratory, but something simpler. I have a magnetic stirrer, some chemicals. Three 3D printers, a laser engraver, a CNC milling machine for milling printed circuit boards with a 0.1 mm cutter. But in the video the laboratory is better and the projects are more interesting 😀
I'm trying to get the fixins to start my mom on tissue culture for Xmas, she was growing a leak suspended by toothpicks at the surface of a jar of water, and my everpresant desire to expand and complicate things lead me to you, the biocoupler kit is on its way, but my budget says the flow hood, stirplates, pipepettel she will have to do with out...Hopfully theres some luck to be had with a primitave go at it.

I flipping love lab equipment. Don't feel bad about falling for the brand new Isotemp. We use them in one of the Micro labs (for college students), and I would have so much trouble denying myself the pretty pretty (except I'm broke). We had a terrible time with our autoclaves not getting up to temp or not staying at temp long enough (while we were waiting on a whole new set of them to be bought and installed), and multiple labs bought Instant Pots as a backup. We still use ours when we have very small things to sterilize and we want to save some energy. (This is a BSL 1-2 lab where we need to sterilize media for bacterial growth and a genetics lab that I think uses agar plates with their C. elegans.)
Is there a reason you use the older "blurple"(purple) growing lights in some areas, rather than using white, full-spectrum throughout? Depending on how precise the pH readings need to be, there are also rolls of pH paper tape(litmus paper) that change color to indicate the pH of the solution. there is a 4th type of sterilizer that is good for anything slightly larger than a loop, and thats an induction sterilizer, they are very fast and zero heat up time
here in asia, i use to try and make a lab but after trying to set it up for a while. that when i realize I don't have enough buget for it even for a super small one as for 1 small machine like sterling machine, and gas autoclave cost like 1 pay check per machine 😢

I make induction heaters for like $30 which are great for scalpels, inoculation loops, syringe needles, etc. never had an issue with contamination

I have a chinesium stir plate from ten years ago that i bought for jump starting yeast cultures for making beer. Still works. No hot plate though.

Is there a reason you use the older "blurple"(purple) growing lights in some areas, rather than using white, full-spectrum throughout? Depending on how precise the pH readings need to be, there are also rolls of pH paper tape(litmus paper) that change color to indicate the pH of the solution. I prefer scalpels. I'm cursed(not literally) and can't use scissors precisely. Love exactoknives and scalpels.
You should check out safety scalpels if you need to make some very tiny specimens or what not. Fisher scientific has a nice one, but a bit pricey! 😝

there is a 4th type of sterilizer that is good for anything slightly larger than a loop, and thats an induction sterilizer, they are very fast and zero heat up time

here in asia, i use to try and make a lab but after trying to set it up for a while. that when i realize I don't have enough buget for it even for a super small one as for 1 small machine like sterling machine, and gas autoclave cost like 1 pay check per machine 😢
