Guile is most likely LeBlanc. Which also has potential for some interesting stories if we get a show there, because LeBlanc is one of the "enemies within" that Swain is talking about. He knows that, and she knows that he knows, so they are publicly working together while trying to have each other assassinated. And it's also implied that they occasionally hook up, Swain calls them "enemies with benefits". As far as political thrillers go, you couldn't do better than Noxus in this universe.
Swain's parents were killed by Leblanc and Leblanc manipulated Swain into thinking someone else killed them. Noxus is a nation torn by differing goals. Swain aims to solidify Noxus' influence across Runeterra through conquest, be it through bond or blood. But LeBlanc has a different goal. She aims to prevent the third coming of Mordekaiser. Back in his living days, Sahn Uzal conquered a large part of Valoran for his religion, only to find out it was all lies. In his second coming, he whispered into the living, tempting foolish sorcerers into bringing him back to the living, now using the name Mordekaiser. He conquered what is currently the size of current Noxus. But he was betrayed, and he allowed them to, in order to rebuild the death realm by using the souls of those he had slain during his revival. Now that his domain is completed, he bides his time for when he can return once again, for he knows his arrival is inevitable. And LeBlanc is finding ways to defeat Mordekaiser by scouring the entire Runeterra, even resorting to using Mordekaiser's former demon servants Tybaulk and Atakhan, although it proved to be difficult to control the both of them.

Alright, so Swain is the "emperor" of Noxus, he's the one who rules, however... He doesn't rule alone. After striking a deal with Raum, the demon of secrets (the demon is his left arm), and gaining all the powers that came with it, he feared that one day he would've lost control and the demon would fully dominate him. So after seizing control of the Empire, he established the Trifarix, the suffix "Tri" meaning three, which gave control over the three different principles of strength to three different peole: Might, represented by Darius Vision, represented by Swain Guile, represented by a yet unnamed pale woman (likely LeBlanc or one of her puppets) He established the Trifarix so that no one men would be able to hold ALL the power, and rule alone. So in short, yes, Swain is effectively the defacto leader/emperor of Noxus, but he still doesn't hold ALL of the power as a failsafe in case the demon was ever to take over, hopefully that clears some questions😊
Actually Swain goes into battle as well, his demon ("Raum") is the demon of secrets, so because of this he can in some way "Predict" what the enemies will do.
