Ha, ha I totally use my insta pot all the time. It works great. I just set the time on the sterilize settings to 1/2 hour. I do media, foil packs, you name it. I did start off with some blank PDA to test how well it worked.
oh man, cutting the saran wrap with pvc pipe cutters is genius. i started buying parafilm because i just got sick of trying to cut wrap rolls

in Mushroomcultivation you let the substrate sit for some houres to incubate what is in there and after this it goes in to the pressure canner, to make sure everythig dies

Hello! Did you try temporary immersion bioreactor? I am looking for the components to diy.
I look through your videos and your content really inspires me and everything looks pretty straightforward, but some of my friends keep telling the way you introduce the whole process of TC is "just for fun" and they mainly point out that having a "DIY" laminar cabinet will drastically lower the overall results in TC. I as a person who's just interested in this sphere, will definitelly have extremely miserable results. Could you please tell if having a "professional" laminar cabinet/autoclave increase the chances? If so how much? Will it require the person to go deeper and deeper into this area to get some visible results? Cause it feels like without having more profound knowledge about this issue, it will waste time and money. I understand that you have really big experience of many years studying and researching and doing TC, but hand on heart will the person "who just started" have sooner results or not, especially having a DIY tools? Thanks)
Hi! I actually just started doing tissue culture in 2022. I have researched the process a lot, but I don't have any formal training or education. Anyone can do tissue culture. Having a laminar flow hood decreases the likelihood of contamination but I have seen many people successfully do tissue culture using a still air box instead of a flow hood.

If I had the space for this, I would blow hundreds in a heartbeat and then spend it again on weird plants. If you take requests / suggestions, Begonia lyallii is a rarely available (but not super rare) terrarium begonia from Madagascar that I'd totally buy so I can stick in my pygmy chameleon tank.
I just started attempting tissue culture with Begonias. Would love an update on how you do yours.
My next vlog will have some begonia updates! I follow the protocol from Plants in Test Tubes for begonias (clean leaf sections in 10% bleach for 10 minutes and use 0.5mg/L BAP + 0.1mg/L NAA for the PGR's)
Can plants be GRAFTED at the tissue culture level?? Like, can u have pink and white variegated monstera in the same dish? What about unrelated plant species?
Do you have any recommendations on a specific desk to buy? The only one I have right now is from Ikea and it has grooves on it which makes it impossible to sterilize.
I used to have a stainless steel table designed to be used in a kitchen. It ended up rusting, so I'm not sure if I would recommend it. Now I use a lab bench with a heat resistant phenolic resin top. They are super expensive (definitely do not buy one new!) but I see them come up for sale from schools / research institutions sometimes for cheap.
For what it's worth I use an instant pot for sterilization of tools, making my own agar dishes, bulk media sterilization, etc. You just have to to run it a bit longer so the material has time to fully sterilize. It has never been a problem. (Literally ever)
(I do mycology and now do some tissue culture with all the same equip)

Between us, I have a cheap electric pressure cooker (no instapot) on the high setting it gets over 15 psi. A large conventional pressure cooker has the benefit of being large.
Can you make a plant from a piece of grapes from the store? Or only from the leaves?
If you found a protocol for grapes, it would definitely specify! I only work with houseplants
Can you do a video that is the whole process from start to finish (initiation to acclimating)? I know it would take months to be ready, but it would be interesting to see the progress with date stamps, etc.

Do you still use fluvial statum and perlite as your substrate? I acclimated some Pluto Philodendrons and creatingthe holes for for their rooted booties was impossible
It's easier if the media is wet. But yes that is what I still use!
What can be used to lift your racks in the PC so high?
I use balled up aluminum foil to lift up the bottom rack

I’m a home mycologist, started in a carpet room with no SAB or flow hood and yes, I’ve set gloves on fire while culturing more than once, I just don’t use gloves no more, and I don’t have carpet now 🎉

What is the largest size glass container Theoretically a tissue culture plant can be kept in? Is there limitations to size? Do you think a one gallon jar would be considered unreasonable? I want the plant to be able to get 8"-10" tall, That way I don't have to mess with them as much...
You could try a container that large but I think it would be much easier for it to get contaminated. I haven't seen a plant grow that large in TC before but could be a fun experiment :)

I think these sterile plants floating in tubes with nutrient solution just look cool af.
or like little self contained plant jars with LEDs that you can put in dark rooms for decoration.
and making it look like interior design of the future 😂

In the cultivation of mushrooms, the community successfully sterilizes @ 12psi for 45 minutes for agar. I'm guessing it would also work here.
I was under the impression that sterilizing at 10-12psi just required longer times?

If you are taking a tissue culture from a plant and your placing it in a soy protein that was genetically modified, does that make the plant genetically modified as well when the new plants are planted.
