Using Agar-agar, malt, and a few other ingredients, this is a very simple and basic agar to use for years. You can then use this agar to pour Petri dishes and slants for cultures.

    I strongly recommend the practice of saving excess grain and bulk-sub water for use with making agar. I have been very pleased with the results of this practice.

    Have you ever heard of foam inserts for those flasks? They are autoclavable and reusable

    Not sure if you covered this but if they are borosilicate flasks they won’t crack you can plunge them straight into an ice bath

    Cant wait to have a go. Buying pressure cookers for this purpose is not easy here in the UK. We have to import them, as soon as I get mine I will be rearing to go with the aid of your amazing tutorials - massive thanks and hugs

      Out of absolute curiosity, why do you have to import them? Government thing, think they are keeping your safe? (Gun control, knife control, pressure cooker control... Mind control)

      @@neogeo8267 Can't speak for the OP, but in my country the issue is that you can't find any decent sized ones. They're all small 4.5 - 6 liters, which fit _maybe_ four 720 ml jars. Preparing any decent amount of grain spawn with a cooker that small is a major pain in the ass as it requires multiple batches.

      @@neogeo8267 theyre probably imported into the US too if theyre made in china. But i think the issue is if there's a supplier that buys in bulk and resells them in your country. I remember watching how theres like only one company now in the US that manufactures enamel dutch ovens and cast iron i think as the video I watched said all the other companies went overseas. I forgot the name cause the product looked good, but ya most production is offshored of most things. Even the US military did a report saying how theyre reliant on china for inputs which is funny. The military is reliant on china for inputs to then face down china haha.

    I'm using my oat soaking water with some peptone and LME in my agar plates. Its working great! I get strong growth within 24 hours!!!

    Add soy/sawdust pellets (or a toothpick, as in another video) for WOOD-loving mushrooms!!!
    e.g., Shiitake, Enoki, Nameko, Maitake, Reishi, Lion's Mane, Pioppino, etc.

    I never had my media glass crack (Especially borosilicate).....and I usually use 40oz beer bottles (not borosilicate) that I toss when I'm done.
    ...but hey, whatever works.

      Thermal shock caused the crack most likely, happens more often on metal tables, simply setting it down on a paper towel can prevent it, edit- later in the video now I'm seeing he said from cooling too fast in the pc, i dont use flasks so I'm able to put more water in so I dont worry about boil over or cracked glass from cooling, cooldown takes longer but it works for me since I pour a lot of plates at once and am able to set mine to hold at ready-to-pour temp so I just do other work and come back later in the day/the next day.
      I like the hotplate idea a lot though, I've never fully trusted microwaved agar despite it being an established practice. Slowly remelting in front of the hood eliminates any (admittedly unfounded) anxiety

    In absence of a hepa filter, how would you recommend to pour the agar to prevent contamination?
    In your previous agar video do you use regular alcohol or isopropyl alcohol to sterilize?

      Isopropyl alcohol. If your using petri dishes a glove box is a good tool or you could use half pint wide mouth mason jars and you can mix the agar normally and pour into jars and then pressure cook. This is assuming you have no flowhood.

    Do you compensate for the soy bean and fuel pellets dust? Working with soy bean pellets comes with more dust than fuel pellets.

    This may have been asked/answered already, but isn't your agar already nutritious enough? Why do you add and what benefits do you have by adding the soybean and pellets?

      The soybeans and pellets are likely the substrate that he is going to eventually grow these cultures on. By adding them to the agar in the beginning, the mushrooms will start acclimating to these Ingredients and developing metabolites to eat them. This helps make the cultures better at colonizing the future substrate. If you intend to use rye or bird seed as a grain, you could also add them to the agar mix for the same reason.

    Is there any benefit in actually coloring the agar, except for it looking fancy and technological? I could think of better contrast in visually inspecting the petris, any thoughts?

      Nope...you covered it all.
      Actually you only need like 4 drops from the bottle (depending on how dark you want) , and sometimes the mycelium will make it disappear.

    How do you get your pellet substrate to 60 % moisture content? Is there a formula at first? I am trying to zero in on 9 lb blocks using 4 lbs pellets and 6 lbs water. Reason for 9 lbers is cause i bought 1000 of the medium bags and anything over 9 lbs makes it harder to mix inoculated substrate. Also seen a video of a guy who puts two agar wedges on the same plate and then does a sectoring transfer from where two rizomorphs join together.

      You just need to do the math.
      You need your 9lb block hydrated @ 60%
      9lb x 60% = 5.4lbs (your dry weight)
      So now you have the dry weight (5.4lbs) which means you'll subtract that from 9lbs and get 3.6lbs (this will need to be the weight of water required).
      5.4lbs dry plus 3.6lbs water = 9lbs for your block. This usually is easier in metric to scale out.
