The essence of the MPN method is to dilute the sample to such a degree that bacteria in the tubes will sometimes but not always contain viable organisms. This is for estimating how many are there in certain volume of water.
Specific amounts were added using micropipette. All the same volume of culture medium but different amount of water sample. The amounts are 10ml, 1 ml and 0.1ml

Thank you Ma'am, now I can answer my activity about bacteriological analysis examination of water...
God bless 😇
Thank you ma'am this video help me a lot in doing my projects and assignment on bacteriology :)

if we're only going to assess the fecal coliform count, do we have to do the completed test still?
If you are only after of total coliform, you may not proceed to completed test.
what did you put inside the durham tube?
Same culture medium, lactose broth
Thankyou so much mam 😊😊
Nice video!
Super 👌 mam
Hindi po ba umaangat yung media sa durham tubes after ma autoclave ?
Paanong umaangat? You mean nagkakaroon ng bubbles after autoclaving? As per experience, hindi naman.
Nice video maam❤
Thanks
mam, first filled with DSLB and SSLB in testube? what's inside of the durahms tube? water sample or media? thats not clear!
The durham tubes contain the same culture medium as with the test tube, dslb and sslb. You just simply put the durham tube in the test tubes inverted.
Thank you mam ❤️
mam can u plzz tell why double standard laactose and why we did dilution of 0.1ml ;1ml and 10 ml ?
The essence of the MPN method is to dilute the sample to such a degree that bacteria in the tubes will sometimes but not always contain viable organisms. This is for estimating how many are there in certain volume of water.
Double strength lactose and single strength lactose. Double strength lactose when you are inoculating bigger volume of water.
Maam where did you get the MPN Table?
From published articles and book by Brown 2005

Welcome. Hope it helps...
please i am not clear to how you added the different dilutions of water to the various test tubes for both the DSLB and SSLB , can you explain
Specific amounts were added using micropipette. All the same volume of culture medium but different amount of water sample. The amounts are 10ml, 1 ml and 0.1ml
ano po yung nilagay sa loob ng durham tubes? salamat
The same culture medium, nutrient broth po
@@jhonamiemabuhay1886 pareho po ng dslb tsaka sslb po yung laman po nun?
