The durham tubes contain the same culture medium as with the test tube, dslb and sslb. You just simply put the durham tube in the test tubes inverted.

The essence of the MPN method is to dilute the sample to such a degree that bacteria in the tubes will sometimes but not always contain viable organisms. This is for estimating how many are there in certain volume of water.

Double strength lactose and single strength lactose. Double strength lactose when you are inoculating bigger volume of water.

Specific amounts were added using micropipette. All the same volume of culture medium but different amount of water sample. The amounts are 10ml, 1 ml and 0.1ml
