    This video demonstrates the streak plate method for isolating a single colony of bacteria from a culture. Proper use of a bunsen burner, inoculation loop, and aseptic technique are shown. This activity is included in the laboratory textbook Biotechnology: A Laboratory Skills Course.

    Very thorough, one comment when sterilizing the loop it is better to start at the hilt and proceed to the loop to reduce aerosolization of bacteria

    When the narrator says rotate 45 degrees, this should be 90 degrees for a quarter of a turn. Other than that, great video

    I am extremely curious who the narrator of these Basic Biotechnology Techniques is. I think I want to be him for Halloween? Can you ask him if that would be okay? He's had a great influence on me and the rest of my Evolution and Genetics High School class. He's my personal hero.

    what would happen if you break the surface of the agar? when I do streak plate I sometimes press the loop hard and it go inside the agar but not all the way..also after each time you flame the loop do you have to let it cool down before streaking it in the plate?

      +BioRadLifeScience some use markers on their plate to draw a line and it will give them 4 surfaces on their plate to streak. is it OK if the loop could come across the like with each streak u do. e.g for the first streak you need to inoculate the loop with the strain..streak it on the first surface then flame it and streak the second surface with the loop coming across the first surface (between the line that separate the first and second surface)...then flame it again and streak the third surface making the loop come across the second and third surface...then for the fourth surface you streak in between the third and fourth..hope you get the picture of what I'm asking..and is it true that the last streak you do should not come across with the first streak you did? thanks for your previous reply

      cool it down first,because the heat can kill your specimen and if you break the agar surface it can give your specimen to growth in colony.

    This will only work on a 100mm plate. It is too many passes for a 60mm one. I am really struggling to get pickable colonies with a 60mm...so little surface area!

      No it’s not visible immediately after streaking.

    Wouldn’t the remaking heat in the tool kill the Bacteria?

      I think that's why he starts the next streak quadrant in the previous one which grabs the bacteria from the previous quadrant and spreads it into the new one.

      the loop is needed to be flame to sterilize it. Yes, you are not wrong, the bacteria will be killed in this process because sterilize is the process of removing any microorganism on or in the metal/object/thing. but the thing is, we need to spread the bacteria on the nutrient agar. if you notice the first process is to dip the the loop on the medium that full of bacteria. from that, he transfer the bacteria by streaking it on the new nutrient agar. that means the bacteria is NOW on the NUTRIENT AGAR and before you streaking the loop again, you need to cool it down for a second. thank you.

      That is why you start a new streak from the previous quad. The goal is to thin out the colonies.

      quadrants means 4 parts of a circle. in a circle which means the petri dish, he streaked 4 quadrants.

    Only problem I can see with this is that he's wearing gloves around a Busen burner, which is actually very dangerous. I'm assuming that those specific gloves aren't flammable but a lot of gloves in a laboratory are and so it would be a good idea to mention this so that students wouldn't repeat
