My mom as well; and she's only in her 60's. I live with her so she doesn't have to go in a home. It's the most evil of diseases. Slow, meticulous, relentless, and ruthless. I wouldn't wish this disease on someone who I absolutely despise. I know exactly what you're going through man. Hardest thing I've ever dealt with in my life; along with the loss of one of my best friends a few years back.

My mother is into the moderate stages of this disease. It's no fun to watch a loved one deteriorate mentally.
What's frustrating is that even if a drug stops or even eliminates plaques and tangles, the damage is already done by the time the dementia is noticed. It's not something that just starts happening and the symptoms crop up in days/weeks/months. Now that I know what to look for, I see that my mother was showing precursor symptoms as much as 10 - 15 years ago.
I’m currently researching and developing drugs for AD and I can say that the general field of autophagy research looks promising for future AD patients.

My 94 year old mother has stage 4/5 Alzheimers and now lives in a Care Home. She was first diagnosed in 2014 with MCI. Mother now relies on a wheeled walking frame and is very much institutionalised. She is not the mother or grandmother we all knew years ago. We lose more of her each time we visit. Her moments of lucidity are infrequent whilst her repetition and confusion are prevalent. Mother knows she will not be coming home and we now face the enormous task of funding her care. Her bungalow and all savings will be used to contribute towards this. If mother had capacity, she would be horrified.

What are the reasons/causes that the Amyloid Precursor Protein is cleaved sequencially by these two enzymes: (Betta) Secretase and (Gamma) Secretase. And makes possible the creation of these molecules principally the Amyloid-(Betta) (A(Betta))?

It's mostly due to some specific inherited genes....and in some cases it is also triggered by aging
Afaik, every protein in our body has a shelf life, before the protein reaches it's end of life it must be broken down, recycled and replaced, this is why the two proteins cleave the Amyloid Precursor Protein so that it can be further broken down and replaced, sometimes due to a malfunction in the recycling stage(or so it is believed) the A-betta proteins fails to recycle properly and over-time it starts to accumulate outside the brain cells like garbage piles.
I am no researcher or doctor, this is what I have read online and since I saw this question I decided to share, hope I helped and hope I'm right xD
Under normal conditions the Amyloid precursor protein is cleaved by alpha secretase and gamma secretase which creates soluble monomers of the beta protein. As they are soluble, they are cleared away by extracellular enzymes. The problem occurs when beta secretase teams up with gamma secretase. There is no explanation as to why the alpha secretase is replaced by beta secretase but certain genes do increase the this.

Yes but I think a certain level of detail has been missed out for the sake of simplicity. The tau proteins are modified by excessive phosphorylation which may be transmissible from one tau to another.
I'm not aware of prions using such mechanisms in their misfolding process but there is much I don't know.

Use that to explain the scenario. They may either help you out, refer you to the other doctors, or they can dismiss the call. Just try. Also, look for forums on the internet.

Is amyloid-β really a target molecule for therapy?
Yes- drug companies are trying to make therapy that will target the secretase enzymes so Aβ formation will be prevented. Other strategies will be to disaggregate amyloid plaques after formation. Also, another strategy could be to immunize people with Aβ antibodies so our immune system is 'taught' to attack these amyloid plaques.
When I first became sick, I started developing short term memory problems. In addition, I'm still considering working with Apple. There are ways to divide distinctive issues between Microsoft and Apple/Google. When it comes to education, it easy enough to share the domestic and international request and load.

My heart goes out to anyone/everyone affected by Alzheimer's/Dementia. A huge shout out to any caregiver doing this alone! I feel your journey. I feel your pain because I've been there. My mom was also diagnosed in her 60s. Feeling absolutely shattered, heartbroken, and drained, working with different physicians across the country with little or no improvement. I was exhausted and overwhelmed to look any further. Well, that is true! :) I was thinking about the way the action potential propagation is depicted like a burning fuse. If we take APs to travel at ~1m/s and to last ~1ms, the active length would be ~1mm. So if the white flash represents depolarization, the whole neuron should light up, not just a tiny part of it. (unless the flash represents only the very start of the AP). I just think it wouldn't harm to represent APs in a less potentially misleading way.

I came here after listening to everywhere at the end of time.
I was diagnosed with renal cell carcinoma (kidney cancer) and Alzheimer’s disease in which I have tried all I could with medical prescription all to no avail.february 1999 and had my left kidney removed at that time. It was determined that no other treatment was needed. This was an incidental finding, seen on an ultrasound to check my gall bladder - I had no signs or symptoms from this tumor. I was followed for 6 years and was deemed cured.
In April 2014, 15 years later, I noticed a lump at the top of my sternum and it turned out to be renal cell carcinoma from the tumor previously removed. I am now Stage 4 as this is a metastasis. The cancer had grown through and broke the bone - I never had any pain or symptoms. I had 10 radiation treatments and was put on oral in August 2014. I started experiencing bone pain in Feb/March 2016 and was given an additional 10 radiation treatments which eliminated the pain. A break in the bone just below the tumor was later identified when the scar tissue developed. I remain on daily oral chemotherapy, I was recently introduced to a Chinese herbalists Dr sale bashiru I contacted him and after necessary questions and forms fill out sent me herbs and oil that cured me of all this diseases within three months of trial.im now symptoms and pain free.here is my email address if you may need help. sarahsmi100@gmail.com

ERROR in this animation. 1:25 shows an action potential spreading in two and going in opposite directions. Action potentials are uni-directional not multi-directional
Actually, AP flow in all directions available to them (like any other neuroelectrical events), which is why they "back-propagate" from their source in the axon initial segment into the soma and dendrites in addition to traveling down the axon. What is represented at ~1:25 is an EPSP and it too flows in both directions so this is not an error.
