So I need to replace these on my car. Can I remove this assembly without any other (mainly drive tube) items being removed? Thanks

Yes I believe you can remove without dropping the drive shaft tube, however, the exhaust and exhaust shields will have to come off to get access up under there and you will need a Lift, laying on your back will add to the drama.

Can this be done without a hydraulic press?

Could be, maybe a good Vise would handle the task?
