I can try to make a compilation of papers I have read overtime (which I think will take more time than I have at the moment) but I worry that the material simply may be overwhelming. For the rRNA and tRNA do they use the intrinsic termination mechanism to undergo termination? As rRNA has a NUT site that blocks the Rho protein, let's say in intrinsic mechanism, the NUT site will be blocked outside of the transcription bubble by the Trigger loop, and the RNA: DNA hybrid is broken by the lid and rudder at the exit of RNA and DNA. Hope to get some feedback 🫶🏻
Why doesn't nus factor prevent secondry structure formation at termination site during rho independent process since nus do it during elongation process...plz explain
NusG (typically) is the anti-terminator/elongation factor. But NusA is a termination factor. So not all Nus factors prevent secondary structures. Also, recent studies show that NusG can also bind to certain sites and stabilize secondary structures and thus, promoting secondary structure formation.
The hairpin forms because there is internal complementarity in the RNA/DNA - i.e. the definition of a hairpin. You don't need external factors to form a hairpin - maybe to stabilize it, but not to form it.
Did you explained the RNA polymerase proofreading mechanisms somewhere... If so please tell me. BTW the videos are amazing, I learned the whole mRNA trasncription from here.. thanks to you I was able to prepare for my MGenetics Quiz.

What I didnt get just yet is how the haripin itself is formed. Are there any factors bending it or is it something else? Anyhow great video.
