This wasn't explicitly said...but i really think the villain was taking vengeance on the antihero for not paying that one time, prepared a evil plan such manipulate his sister to open the store next to his, and break them apart... but then the sister turned into a donkey (probably her ability) and then replaced the donkey...
The second song the villain says someone didn't pay him once, and that made him want vengeance... then we see hoe for almost not aparent reason he made the two brothers break apart, but I really think this was his vengeance plan
