The True Story of David Sinclair's Longevity Lie

  • The Harvard geneticist sold a fake miracle pill to GlaxoSmithKline for $720,000,000, and now wants the world to believe that he has discovered a new immortality molecule.
    Everyone wants to live a long and healthy life. No one wants to die. The oldest scam in history is the longevity lie. The first writing recorded on Sumerian clay tablets recounts the story of Gilgamesh’s failed quest to bring his friend back from the dead. The immutable fact of mortality has dogged human kind since its very beginning. And yet every age has brought with it its own crop of magicians, alchemists and scientists promising eternal life. Their pitch is always the same: everyone who came before them was a charlatan, but they have the secret sauce.
    The most famous longevity grifter of our age is no different. If you’ve ever heard a news story that a glass of red wine might make you live longer, it was because of his groundbreaking research. Harvard geneticist David Sinclair is one of the most decorated scientists on the planet. He’s listed as an author on more than 500 papers, his work has been cited more than 96,000 times and he holds 50 patents. He was the editor of the journal Aging.
    Resume aside, David Sinclair is no different than any other health grifter throughout the ages, and great fortunes have been squandered in pursuit of his “science.” In this week’s video I dive into his 25 year history of scientific mistakes, lies and fraud. I show how he used disproven research on the chemical “resveratrol” to sell a best selling book and, ultimately, a company to the pharmaceutical drug maker GlaxoSmithKlein for $720,000,000. Two years after the sale, the research was proven to not work, and Sinclair became one of the richest scientists in America. Now, ten years after that work fell apart, Sinclair is at it again selling the idea of a new immortality molecule called NMN.
    This video took me three weeks to put together, but it was worth the wait.
    CORRECTIONS: Robert Sclafani, Ph.D. is a professor emeritus at University of Colorado School of Medicine in Aurora, not CU-Boulder as stated in this video. Additionally, his meeting with David Sinclair and the member of the Nobel Prize committee occurred in Colorado, not Europe. More about Sclafani here: medschool.cuan...

    The best longevity ideas so far are getting enough sleep and regular exercise, eating healthy food and drinking water, avoiding cigarettes, alcohol and drugs, driving responsibly, avoiding crime so you stay out of prison, avoiding arguments that lead to fights, worrying creatively rather than extensively, doing creative activities and helping others while not getting used, spending time with friends, living a meaningful life, feeling inspired by something or someone worthwhile, being interested in a fair few things AND it's also important to have a good superannuation plan. Start paying into it while you're young. Keep paying into it until you retire and then you can have quality of life in your old age.

    I am 61 years old and when I take NMN I walk 5 miles a day, when I stop taking it I stop taking walks. A friend of mine had 4 strokes and was laid up in a chair for weeks. I sent him some NMN and the next day he was out puttering around in the yard. Hasn't has a stroke since. David Sinclair's NMN is one of the only supplements with a noticeable effect. You can't debunk NMN for me. I would love to see the science on why NMN doesn't work. And why is the US Military doing studies on NMN as we speak. Why is NMN present in every cell in our body? A few jealous and petty doctors who prescribe poison every day with hundreds of side effects are mad they have not gotten rich on something they discovered. Resveratrol may not be what it was hyped to be but it is still a player in longevity even as a possible antagonist. Quack, jealous doctors getting rich off the backs of real scientists, that is the problem here...

    1) I don't like the strawmanning going on in the video introduction and description ("wants the world to believe that he has discovered a new immortality molecule"). Sinclair never claimed that some single molecule/drug he has discovered confers immortality, that's a flat-out misrepresentation.
    2) From the video description: "I show how he used disproven research on the chemical “resveratrol” to sell a best selling book and, ultimately, a company to the pharmaceutical drug maker GlaxoSmithKlein for $720,000,000." - It's the other way around. Sinclair sold the company in 2007. The book only came out in 2019.
    3) The copious use of emotional appeals - words like "grift" and "lie" - from the outset, ironically, make you sound yellow-pressey. Objective reporting should be more aloof, or at least save terms of this sort for the end, when you've made your case and their use is justified.
    I've been meaning to read your book "Red Market" for a while now, but given your apparently sloppy and tendentious approach to reporting, maybe I won't.
    P.S. I'm not a Sinclair fan. He's recently been ostracized by his peers, ostensibly deservedly. But his science is still possibly better than your journalism.

      I tend to agree with you.
      By constantly using words like 'grift', the video is priming the audience's brains to draw the conclusion that he wants it to draw.
      In the end, all he has is basically one professor vouching for his claims that Sinclair is a fraud.
      Also, the resveratrol claim isn't the only thing Sinclair promotes. He also promotes intermittent fasting and cold showers, which are ideas that earn him zero pennies. This video doesn't address the science behind that.
      Finally, for a video that purports to be a piece of journalism, that final bit on the Nobel prize rings like silly 'high school mean girl' type gossip.
      The presenter smiles like he's enjoying this bashing a bit too much.

    • @MichaelAachen
      @MichaelAachen 3 หลายเดือนก่อน +10

      @@studentofspacetime 1) "By constantly using words like 'grift', the video is priming the audience's brains to draw the conclusion that he wants it to draw." - Yes, and I wouldn't really harp on this point if Scott Carney was your average clickbaity TH-camr not pretending to do serious journalism, - excessive emotional appeals would be par for the course, - but Mr. Carney's credentials as per his Website bio, etc. seem to establish him as a (near-)professional journalist, who just happens to have a TH-cam channel. Thus I had higher expectations.
      2) "that final bit on the Nobel prize rings like silly 'high school mean girl' type gossip." - Oh yeah. Sinclair's networking - which I would expect all sociable scientists are doing - is painted in the most unfavorable light possible. Indeed, with a kind of mocking, slightly childish glee, as you imply.
      But I honestly had so much issues with this video pertaining to its factual content. Writing out a fully detailed critique would just take too long, and I can't even be certain the author would see it. Just a couple of examples:
      - “The Sir2 theory was junk.” (Mr. Carney) - Not so fast, it’s not that simple. A scientific article from 2014 called “The controversial world of sirtuins” and published in “Drug Discovery Today: Technologies” provides a nice and relatively comprehensive overview of the historical studies of sirtuins and the controversies around them. The conclusion? “Sirtuins (can) be therapeutic targets in future medicine… Despite so many controversies, mammalian sirtuins have been clearly shown as a class of critical factors regulating many cellular processes, playing important functions in diverse tissues and systems… However, the complexity of activities of sirtuins, and their widespread roles and activities, increases the difficulties in determining how best to modulate them therapeutically.” The upshot is that a) sirtuins are definitely a key player in human biochemistry, b) we don’t understand either it or them well-enough yet to make conclusive statements or develop targeted therapies.
      Incidentally, the article’s author, a Mr. Dang, summarizes the goings-on around that study by Matt Kaeberlein mentioned in the video. Sure enough, several studies in Europe didn’t replicate its results. But then another study did achieve somewhat similar results. The experimenters on both sides are still arguing about the details, and the issue is not quite resolved.
      - Re: the claim that Sinclair’s blocked "all the companies that were selling NMN" from the market. - However, the very document Mr. Carney displays on-screen in connection with this claim states clearly that this does not apply to the companies who’d already been selling such products before Sinclair’s team filed their paperwork. Also, a perfunctory search on Wiki shows that companies had been selling NAD activators (which is what NMN is) as supplements well before Sinclair put the spotlight on NMN.

She didn't try to end her life or anything, just complained a lot because what she spent her time on was reading and doing needle point so that was lost. The only thing I remember her doing for longevity is drinking a lot of hot water. At least once a day she would put a teaspoon of honey and a teaspoon of apple cider vinegar in it. She also ate a lot of grease, she like to wipe out a cast iron skillet which had fried a bunch of meat in it, with a piece of bread and then she would eat the bread. She also put gravy on everything. When he was leaving the lab to start his lab at Harvard, he and Lenny (Guarante, my graduate advisor) agreed on which projects he would take from the lab, a normal part of the process when starting a new lab, and I don't think Sir2 was supposed to be part of the mix.
      As for Matt Kaberlein, from a personal standpoint, I was not particularly close to him even though we overlapped for a fair amount of time. I was actually much friendlier with Dave Sinclair, and we would frequently go out as part of a group for a drink after work. That being said, Matt was much more rigorous than Dave in his science and is typically much more careful in his claims now.
      Both Matt and Dave are extremely smart and extremely driven. This is why they have been so successful. These things do the heavy lifting, however, some people are lazy and would like to just take a pill. They can be gullible to snake oil salesmen. While this video might suggest otherwise, David Sinclair is still very much active in longevity research. Contrary to the claim that he has been "exposed" and is no longer contributing to science, Sinclair continues to work on cutting-edge projects, including epigenetic reprogramming, which shows promise in reversing aspects of aging. The field of longevity is far from stalled; in fact, advancements in AI are accelerating research, allowing us to explore new possibilities more rapidly than ever before.Rather than dismissing the entire field because of perceived failings by prominent figures, it’s crucial to recognize that the pursuit of understanding and potentially reversing aging is a noble and ongoing endeavor. Proper breathing can be extremely effective for both physical and mental health. On a physical level this will result in richer oxygenation of the blood which will result in many health benefits and perhaps even a longer life. But the true power of the breath comes into play when it is used to achieve a calmer and more centered being. You become better equipped to deal with life's difficulties and stressful situations. It can also be helpful in times of extreme grief and bereavement. The book “the full breath” of James Francis explains all this and also explains in simple terms the concept of spiritual development and enlightenment. One of the things that can ruin our health and our lives more than anything else is stress and mental distress. If you know someone who is suffering in life or is in great difficulty, this book could help them fight their way out and become happy and balanced again.

    • @Fomites
      @Fomites 3 หลายเดือนก่อน

      "Proper breathing" may have health benefits - but not from extra oxygenation of blood - this is because of the nature of hemoglobin and the oxygen association curve it produces. Animals are pretty much at 97-98% hemoglobin oxygenation all the time regardless. All "proper" (in other words harder breathing or hyperventilation) will do is cause mild alkalosis from CO2 loss. Oxygen is also a toxin - it's essential but still a toxin and thus we have extensive mechanisms to mitigate its toxicity.

I have never felt better and I have seen substantial differences! I don’t take a lot, but add it in 5 out of the 7 days. My husband was skeptical, but I had him take the supplement stack for 2 weeks and he gained his energy back and much less pain in his joints. Problem is the issue with the lab artifact fluorescent molecule being needed to activate sirt1, was solved in a subsequent experiment by showing that certain amino acids could substitute for the fluorescent molecule and allow activation of sirt1 by resveratrol in their presence.
    Not only has resveratrol lengthened the lifespan of over half a dozen species, iirc, including vertebrates and mammals. But in vitro it was shown to turn old human cells young and elongating their telomeres and restore youthful phenotype.
    Why did resveratrol fail in longer lived rodents, unlike all the short species it worked on? Could be the high cancer rates of rodents, maybe it doesn't stop cancer from limiting their lifespan. Or could be age related NAD+ decline. It remains unknown why it failed in this case.
    That said the NMN move by Sinclair was scummy. But his cancelation is nothing more than baseless hit pieces. And yes these are effective. You call someone's job and make up stuff, and people can get fired for made up stuff.
    As for vaccines, covid vaccine was unlike any that came before. In a few months it exceeds the reports of side effects of all other vaccines combined in many decades combined. And this was despite most reports being by healthcare professionals, and due to politicized nature most being chastised or advised not to report or blame the vaccine. When canada offers euthanasia to those who've been made disabled by the covid vaccine, and when the poster boy child used to advertise it falling dead. It is not good. And no not all batches showed significant side effects, only some batches did, often killing or maiming within minutes or hours.

      Finally, someone who actually knows what they are talking about.
      It's very easy to do a hit piece and very difficult to provide an accurate picture, because it's often less sensational and more filled with boring details, and there's no clear villain in the end.
      To me it looks like a grifter inception: David seems to have wanted to monetize NMN at the expense of the community, and now TH-camrs monetize David for views. Now we need to go deeper and have TH-camrs expose those TH-camrs who expose David, and everywhere in the chain the titles should get more sensational and the picture portrayed more inaccurate to get more views.

While on stage, I asked “who wants to live forever?“ Entire audience went crazy. “We do!” They all shouted. “Why?” I yelled back. This is part of the scientific process. Theories are put forward, they get challenged and are either refuted or they persist. Confusion results from the business influence which is quick to grasp a marketable magic bullet. However, research continues and accumulates valid scraps of bona fide evidence toward an ultimate goal. Read objectively through this process.

    Average life expectancy has been *decreasing* in the US; snake oil and moonshots are a distraction from the actual life-extending solutions we could implement like universal healthcare

      It decreased temporary during covid. It's probably going to significantly increase due to drugs like GLP-1 analogues having anti-diabetes, anti-obesity, anti-cancer, anti-atherosclerosis, etc effects. These drugs show the path forward by using drugs to modify human biology.

    Would be nice if ppl would be as upset by the fact that 90% of what is sold in grocery stores is unhealthy, that in every tv program, like sitcoms, talkshows etc all our children, well and everyone basically, is being told in a sneaky way to drink alcohol, smoke and eat unhealthy products (often by product placement) and use stuff like Botox to look a certain way. Seems totally socially acceptable. Would be great if investigators look into that and explain to ppl how they are being played.

    I see all these videos coming out retracting all the scientific data from a few years ago. Intermediate fasting, carnivore and keto diets were the be all and end all for superior health and longevity. Now there are videos stating these findings are backfiring for a lot of their youtube followers. I do better on low carb but not the extreme keto or carnivore diets. I have noticed if people have health issues and try an extreme diet then they feel better because they ditched the sad diet. But after many months or years they have to add in some healthy carbs to feel better. That is what I have noticed with all the youtube health gurus. They are fun to watch but I don't take their advice because I know it constantly changes.

    I remember saying under a lex fridman video that him saying pathways in organisms that are separated by millions of years in evolution was a stretch and should be taken with a pinch of salt based on my knowledge from my own PhD and a faculty member of my former department who also worked on longevity. By God, did his fanboys rounded in and attacked me for that statement, it was clear when I tried to engage with them they had little to no knowledge of the basis of what he was trying to say but were wedded to the story of living longer than he was spinning. No doubt he's a smart guy but when he's disseminating bad information it for me promotes scepticism from the general public towards science.

    I read this story about a supercentenarian that lived to 120. She smoked until she was 115. The role of genetics cannot be underestimated

      Outliers always exist. However playing percentages and risk, that is not likely to work out for most people. Genetics mean something but you'd be surprised how much you can fight genetics with exercise and diet.

      We don't judge on outliers. Smoking is causally linked to cancer, for every smoker who makes it over 100 probably about 50 die before 65. Everything I hear suggests that the role of genetics is largely overstated.

    • @jtthoma5
      @jtthoma5 3 หลายเดือนก่อน +3

      @@GraftonPrimaryvideo smoking is terrible for you, I would never condone it. What I'm saying is that there must be very strong genetic factors involved with those outliers, because not all of them lived super-healthy lives (like smoking for a...century). The average person should absolutely have good diet, exercise & sleep habits. But I think as a society we are too obsessed with living to these old ages, and then we use these "genetic outliers"/supercentenarians as examples of what average people should strive for in terms of longevity, which is sort of misleading. As a culture, we should be valorizing individuals that had a high quality of life right up until the end, in my opinion.
