There should be an issue here if I understand well. At 3', during the example, the added nucleotides are in the order : G A T C A G A T etc... However when the second template is "GC", dATP is added, and nothing happens for template 2, only template 1 gets light. But then there is dGTP then an other dATP, WHICH THIS TIME CAUSES LIGHT. Why does the first dATP did not cause light in template 2 ? Just a little error on the video or am I missing something ? Great explanation otherwise

How do u know the specific sequence?
The dNTPs can be matching the nucleotides in the template strands randomly..
They have to be added in sequence, also, it's the phosphodiester bonding that releases pyrophosphate not hydrogen bonding.
