    Can we use the hot plate instead of a digester for the digestion of the dry microbial sample? can this method be incorporated for liquid samples?

      Yes. You can use Hot Plate to digest your dry microbial sample. This method is also applicable for liquid sample.

    Interesting video, MicroChem
    1. Do you have reference method for the procedure? Perhaps by ASTM or the likes? Would appreciate if you can mention this in description.
    2. Suppose the sample is a representative of larger material, is there any requirements for the sample? Particle size? Homogeneity?
    3. I noticed there are two oxidizing agents used i.e nitric acid and perchloric acid. Is there any reason why? Could we make do with only nitric acid? Would appreciate if you could explain the particular reaction between each chemicals with the element of interest (maybe As or Pb?)
    Anyway, looking forward for the Fe by AAS video.

      Answer:
      1. We developed this method at our lab by combining several methods. So no official reference method for this.
      2.Sample should be crushed and blended and homogeneous portion of sample should be taken for the test.
      3. Perchloric acid helps to digest sample properly. So you must use Nitric acid along with the perchloric acid to digest your sample properly.
      *We will make video for Fe analysis by AAS later. Stay with us.

    Hello. I have 3 questions. I hope you guys respond to me.
    1. Is there any difference in procedure when we use this wet digestion method for heavy metals of fish flesh samples?
    2. Incase, how will we dry the fish samples? How many grams we will be needing?
    3. Is this wet digestion can detect various metals after AAS? Or is this method only specific with limited metals?

    Thank you for the explanatory video. When you left the flasks in fume hood to digest the sample in ambient temperature for 24hr do you also left the fume hood power on?

    Which standard range (like 1 ppm, 5ppm, 10 ppm) should be calibrate for higher concentration of element of choice???
    For instance 1,00,000 ppm.

      Prepare calibration curve using 1ppm, 2ppm & 4ppm....not more than that. Dilute the sample to suitable dilution and analyze.

    Heavy Metal content in food sample can be analysed by both way i.e dry ashing or wet digestion.

      Wet digestion is the best as it has low possiblity of metal loss.

      Thats a different thing. You can analyse using the testing kit. But analysis using AAS provides more accurate test results.

    Good morning. I did a wet digestion using this procedure in order to dose aluminium by AAS. In the end I obtained a clear sample after filtration and dilution with pure water but after 24 hours, the sample becomes cloudy (white). I don't know the cause. Is it normal? I have never do or see wet digestion

      Meenakshi Trilokia, yes. You can digest on heating mantle. But you should be careful to avoid dryness of some part of the bottom surface of a conical flask or beaker. That may lead to sample loss.
      Round bottom flask using is more desirable for this purpose.

    Dear MicroChem's Experiment team, I have a question and need help: I have weighed 1.0153g of a sample and digested with 20 ml of acid mixture, then after digestion, distilled water water added to make up the volume to 250 ml. Then 25 ml of the sample solution was taken and titrated against 0.01 N EDTA and 0.2 ml of EDTA was used to get the end point, to calculate the Calcium content and the formula I should use

      There are some other methods where HCl & H2SO4 is used. But We developed this method using HNO3 combined with Perchloric Acid and found best digestibility of HNO3. Thats why HNO3 is used for the digestion.

      You can prepare urine sample in the same way described in this video. But take 10ml of urine sample in the flask instead of 2g. Rest of the procedure will be unchanged.

      Geranally sample is a complex organic compounds. In the digestion process, each of the complex bonds are broken down in the presence of acid, heat and sometime catalyst. When all of the bonds are broken down, all of the minerals and heavy metals are released which can be quantified using AAS.

      Glass beads are used as an anti-bumping agents. Glass beads does not interfare the chemical reactions but prevents bumping the acid solution during heating at higj temperature.

    How can you throw Acid like that, in the sink as well? It's bad for your pipe and the environment. I appreciate your video, but you must keep every aspect, especially health and safety issues

      You are right but It is not possible to maintain everything (e.g: disposal of acid) in the video within a very short time. Because our main focus is to present the test method appropriately. Some times we pipette concentrated acids in the open place, without using the fume hood. We did like that to just shorten the video length. So please maintain health hazard issues at you laboratory while following my video.

    Can I digest the fish gills and liver with the same method using a hotplate?

      Saeed Noori, yes, you can apply the same method. But you should use a digester because fish gills & liver may take longer period of time to digest and in hot plate technique nitric acid removes rapidly. So if you use hot plate then you have to add addition 10ml of nitric acid in sample and blank beakers in the middle of digestion period to prevent rapid dryness of the beakers.
