    Hmmm, I think I have the same problem. How long do you store the sample in the freezer after diluting in 1 to 10 water?

      We usually do it overnight because that's convenient, but if you're working with 1-2mL of liquid, it's similar to an ice cube size, and it'd take ~1-2h to freeze completely. Add a hack: you could separate your DNA extract into aliquots of 0.5mL in small tubes so they freeze faster.

      @@BentoLab By the way so we need to make it solid frozen? Because in my lab we just dip in ice or put in fridge (not frozen)

      @@htrr23 You'd want to aim for frozen solid. The freezing step helps deal with the PCR inhibitors like proteases in the solution - they defrost more slowly that DNA, so it's a quick/cheap fix for removing them without using a column, or a more intense DNA extraction method.
      It's like the reverse of how you need to fully defrost enzymes before use.
