Happy New Year! THX for the video. I don't get the bigger picture. I'm just new to this. You said the Halo around your culture is an indicator for bacterial growth on the plate? That would be a nice detail I should keep in mind.
You need to have the blade hot and turn it sideways to carve a gap with the broad side of the blade. It needs to be a clear gap all the way to the bare Petri dish.
You can really use either. The meat will speed up the growth needed to achieve the hopefully clean transfer. But personally I prefer to make that transfer to water agar..then transfer one more time after that back to MEA
@@RealSimpleMushrooms thanks for the response! i put it back to water agar this morning , but i find the growth incredibly slow and very fine strands ... I will try both these methods on water next .... i was on T6 and it was making me crazy with the aseptic technique .....i was considering antibiotic agar , but i would rather avoid if possible
When I put something to water agar usually it will go rhizo (but be very faint) and as soon as it shoots off from the transfer I’ll grab a piece and toss it back onto MEA

Not 100% of the time but it is an indicator
Is the trench just a cut in the agar or does there need to be a small gap? I find it hard to do anything other than a clean cut.
