Sorry I have been a little quiet these days, I've been busy with PhD applications and work, but I'm back today with another vlog (festive edition)!
I can't believe it's December and already time to whip out my puffiest winter coat. If it's getting cold where you are, I hope you're dressing warm and keeping cosy 🧣
This week in the lab was relatively chill as we just did a major transfection and mostly waiting for our embryonic stem cells to grow. In the meantime, I'm preparing the next construct to transfect (which isn't mega urgent right now). I am mostly doing microbiology work, so plasmid extractions, picking colonies and culturing E. coli. With the plasmids I have prepped, I am doing long-range PCRs to check that the construct is intact.
At the end of the week, I headed to London and had the most amazing time at Winter Wonderland 🎡 Of course, any time in London is a food heaven. But the main reason I made the journey down was because I booked tickets to the musical - Wicked 🧙♀️🔮 (they were on Black Friday sale!) It was so so good I can't stop thinking about it or playing the Broadway soundtrack 💚

Uma coisa que eu acho diferente é que vcs fazem PCR fora do fluxo. Aqui eu faço as minhas todas dentro de fluxo laminar. E tem 3 salas separadas. Uma para montagem da PCR ( reagentes) uma pra colocar o DNA na reação de PCR e uma pra correr. Usamos a estante prós tubos congelada assim mantemos melhor os reagentes durante as montagens de PCR
Yes that's true, I think many labs would do it like you, but we only have one communal one shared between multiple labs and since I do lots of PCRs, it's easier to do it on the bench! Ofc I make sure to clean the surface thoroughly before starting.

This really motivates me to keep making videos, thank you 😊 I graduated with a 1st class (~70%) in undergrad and Distinction (~70%) for my Master's.
One major piece of advice I'd tell myself is to be less of a perfectionist when it comes to taking notes and revising for exams. University exams do have knowledge based questions but I'd say it's more important to understand the principles and how things tie together. I would've focused more time on linking information tgt using mind maps and also actively recalling topics I was worse at rather than going through everything in sequential order - it's pointless to revise something I already know 5 times 😂

I did a placement this summer and really struggles with calculations like the master mix I worked under a postdoc in a base editing project which consisted of many cloning & it involved so much calculations and conversions does this get easier the more you work in the lab
Oh it definitely gets easier with time and practice! My first few times doing all sorts of calculations whether it's serial dilutions, master mixes, calculating reagent volumes for cloning/Gibson assembly, were all confusing. Once I started to get the hang of it and understand the role of each reagent in the process, it starts to make so much more logical sense!

No I do not think it's necessary, or at least I didn't need to to do microbiology work in the lab. And thank you!! 😊
