How do you deal with too much condensation and plant vitrification?
We recommend measuring & adjusting the media pH after adding supplements, including gelling agents. Agar can have a wide pH range. Sodium hydroxide and other bases are used to increase the gel strength of some agars during their manufacture; this can result in a pH range from slightly acidic to basic (pH 9+). It is for this reason that we recommend adding it before checking the pH of your medium.

@Gemthermakleingk Yes, this is done intentionally with polypropylene containers like these. The lids seal tightly which results in deformed containers when the pressure drops at the end of the autoclave cycle. This is circumvented by leaving the lids slightly ajar and then closing them when the autoclave is opened at the end of the cycle. This is not a problem with baby food jars using our PhytaCaps or Magenta B-Caps.

If you’re using an agar with a high pH and adding it after adjusting the pH of your media then it could cause your media’s pH to increase without your knowledge. In many cases this may have negligible impact on your cultures… unless you’re culturing plant tissues where the pH needs to be more tightly controlled.

Your pH electrode will be screwed if you adjust the pH after adding agar. I always add the agar separately to the medium after taking pH. Been doing this since 12 yrs now and never had any problems. :)

On the contrary, as long as the agar is not melted it will have little effect on your electrode. Actually, measuring the pH of media containing proteinaceous components (e.g., peptones, typtone, casein hydrolysate) is more likely to coat your electrode and reduce its sensitivity.

Most plant growth regulators are routinely autoclaved in the medium, especially the stable ones such as BA & NAA. Some are heat labile and should be filter sterilized and added after the medium is autoclaved and cooled a while. Our "Plant Growth Regulators, Part II" document on the Technical Info page on our web site offers guidelines for whether the plant growth regulator can be autoclaved in the medium or filter sterilized.
