- Dynamic motion during loading and activation of the yeast Replisome

Nynke Dekker describes her work on the DNA replication in yeast at the single molecule level. She used optical tweezers to trap DNA between the beads and then visualizes assembly of the fluorescently labelled replication proteins on the DNA. Alternatively, she captures pre-assembled DNA-protein complexes between the beads. This has enabled quantitation of both binding and diffusion of proteins on DNA. This has led to the first successful analysis of CMG motion on DNA and the effect of nucleotide state on this process.
