    2024 Introduction to Biophysical Techniques
    The LMB Mass Spectrometry and Proteomics Facility houses a wide range of mass spectrometers enabling biological proteomics and structural proteomics. Biological proteomics involves the identification of proteins and their interactions and the comparative proteomics of cells and tissues. In structural proteomics purified proteins or their complexes are investigated by hydrogen-deuterium exchange or cross-linking mass spectrometry.
    In this video, Cat begins with the question: what does a mass spectrometer measure and what are its mains components? She describes how samples are ionized, mass is analysed and detected by the mass spectrometer. She explains why mass spectrometers are often coupled to hplcs for sample preparation. She then follows with a detailed description of approaches used in the facility for biological mass spectrometry for proteomics including sample requirements, workflows and labelling strategies for quantification. In addition, she shows different data acquisitions methods for label-free quantification. Lastly, she shows approaches for structural proteomics via cross-linking or by hydrogen-deuterium exchange.
