This is the Edinburgh Comedy festival, it's a notoriously difficult crowd, if they don't like you you'll know it. There's been bigger comedians than him get booed off stage and never want to go back. As pretentious as it is to say the people in the crowd will have forgotten more about comedy acts than we will ever know, they're hanging on his words and don't miss a joke, it's pretty respectful to him!
He played it well, he knew no one would do the call/response so he worked it into awkwardness, hence him subtlety saying "Oh booooo...." and played off it. Clever.
It's actuslly part of the bit! He intentionally doesn't prep them it reinforces the entire songs delusional narrative that there are these women craving him
