    I just found your channel, i wish found early(my grade is not that great), this video makes it easier to understand the procedure since things have switched online and we have no Labs in person but have online work

    Thank you for your guidelines, however I'm not sure I saw when you added the eluting solution (probably the camera didn't capture it or my eyes are playing with me). Would like to know how many mls or drops of the eluting solution you added. Thank you

      The last wash is tested parallel to the eluate (as if it was the patient's plasma) using the antibody identification panel. The idea is that there should only be an antibody in the eluate and not the last wash.
      The eluate should contain the antibody taken off the patient's RBCs. It is important to identify this antibody so as to ensure safe transfusions in the future.
      The last wash should not contain any antibody. If there is a positive reaction to any of the reagent RBCs in the antibody identification panel it could mean a couple of things. The most likely cause would be the wash steps were not performed correctly and there are still some residual unbound antibodies floating around in the last wash. The other scenario is that the working wash solution liberated some of the antibodies from the patient's RBCs (which is not desirable). If this is the case, you might see a mirroring of a reaction pattern with the eluate or negative reactions from the eluate. In any case, you would need to perform the elution and antibody identification panel over again.

      so 50ul of last wash and 25ul of eluate or the other way around? because for antibody Id I will have to use Panel A 50 ul of Rbcs and 25 ul of eluate. I would really appreciate your help.

      Using the tube method, you would have a set of tubes which would test the eluate against the reagent RBCs. At the same time, you would have another set of tubes which would be testing the last wash against the reagent RBCs. So, if you have a 10 cell panel you would have 10 tubes testing the eluate and 10 tubes testing the last wash (20 total).
      You would treat the procedure the same as if you were doing an antibody identification panel with a patient specimen (1 drop of reagent cells and 2 drops of eluate or last wash in place of the plasma).
