No, but if that accessory you are checking only has power when the key is on, then you need to check with the key on. Does that make sense? It really depends on how they are wired. Some things are fused after the ignition "ON". For those things the key would need to be on to check power to the fuse. If you're not sure you can just leave the key ON while probing the fuses.
