Nicotinamide (NAM) from food or supplements can enter the cell through passive diffusion since it is a uncharged, non-polar molecule. Charged and polar molecules (like NAD+) cannot pass through the phospholipid bilayer. This is why you can only increase internal NAD through the use of precursors or by optimizing the salvage pathway.
I take metformin for longevity (I am not diabetic). I can also obtain NAD+ injectable from my metformin supplier. NAD+ increases ATP whereas Metformin reduces the conversion of nutrients to ATP. So my question is, is NAD+ counterproductive to the benefits of metformin? Would it be unwise to take both?
