Basically, crushing help to make smaller pieces, break cell-cell bonding and cell wall, detergent breaks cell membrane and nuclear membrane, salt separates proteins that are attached to DNA, and ethanol help to visualize DNA by making it insoluble (DNA is normally soluble in water) and finally the DNA here is seen actually lots of double helices strand are attached together (not a single double helix). a single strand can be seen in a microscope only. However the video is informative, you have prepared it nicely. Thanks

How to confirm the extracted one is DNA? Is there any simple methods to confirm?

@@gopinaththiru1238 By using spectrophotometer we check the peak absorbance value. It should be around 260nm.

mix the DNA of mustard seeds with jack fruit or fruit which produced more seed so we got more oil
like we mix the DNA of oil plants across the big trees like pipal tree or bargad tree or any large tree

Sir this is incomplete experiment. It may also contain many other macromolecules like proteins and RNA. In order to remove these macromolecules we have to add enzymes like protease and RNase to get them dissolve and get purified DNA.

The initial sample you have taken which you have taken are too little and in that amount of sample hod did you get that much of DNA. Definitely it will come in nanogram. I think your DNA got contaminated with protein and RNA.
