ive germinated a dry tissue of a over 5 years old (fridge stored) specimen of the Gills on WaterAgar (simple Agarrecipe without the sugar - Agaragar + Water). There are millions of spores on the tinyest pcs of gills and the chance that just 2 of them come back to life is extremely high. And it worked like a charm - muchlove 🍄

I also used Wateragar (a Agarrecipe without the nutritionparts - just agar and water). As i just wantet the spores to germinate i think the spores dont need so much water so rehydration is no need at this point :)

Would it not be beneficial to run the dried fruit under a UV lamp in order to destroy bacteria prior to completing the process? At least that way you can assure the outer surface of the fruiting body is bacteria free.
I know this sounds crazy but I just did this, i reconstituted the caps and after I extracted spores I threw the leftover caps in my kitchen scraps which had some uneaten noodles, this was unintentional but when it was time to dump my table scraps later that week into my compost bin, I saw the caps and the noodles were completely covered in mycelium so I tried collecting the “hairs” and put them in a jar with sterile water and honey and used it as an inoculant a couple weeks later, it was the most unsanitary thing to collect the mycelium from my compost pile but it worked out perfectly, apparently noodles work for growing mycelium maybe not the best thing
I’ve never grown mushrooms in my life but I recently picked up a quarter of golden teachers. So I bowled a jar of corn syrup and osmosis water then crushed a dried cap into powder and put it in the jar. Do you think that it will grow mycelium? I have no idea what I’m doing all I know is I gotta put it in grain after and let it colonize
I have done this, but I always assumed I was just germinating some spores - even mushrooms that don't seem to drop spores can still have spores in/on their gills. I think you would need a much more convincing/controlled experiment to confirm you are actually reanimating mycelium.

My only objection with this exact method is that when the gills hit the water, whatever spores are left on them can lift and spread around the rest of the tissue. What if I'll place pieces of dry mushroom in a 3 percent solution of hydrogen peroxide for 1 minute, then rinse in sterile water. and put in sterile water for 12 hours, then rinse again in hydrogen peroxide and then rinse and put in agar?
Unfortunately it doesnt matter much about taking a core sample. Youll still get a few duds in regards to sterilization but i find my favorite way is to use the underside of the mushroom cap if i have access to it. Swabbing it for spores usually does the trick for me and i find that i get less contam from the swab method and can get more plates out of the spores. Maybe ive just been lucky though.

I'm from Vietnam and I have a question I hope you can answer: is there a way to disinfect the fungal spores to get rid of mold and bacteria before inoculating the spores into the petri agar? eg: can i use 90 degree alcohol or brine to soak the mushroom spores, will soaking the mushroom spores in it damage the spores? i have tried putting spores in agar but most of them are contaminated even though i checked my agar is completely sterile... Yeah antibiotic or H2o2 agar...but really u cld improve your chances by taking your tissue from the internal tissue of your specimen....as per usual tissue transfer......inside stem-base or inside stem an cap just above the gills.....but I WLD recommend the inside of the bulbous stem base🎉 BooooM

Hey willy just skip the water step, dries fruit will do the same which will cut out the extra 24 hrs, also the age of the fruit shouldn't matter, I've done this with a specimen over 2 yrs old I had when in Mexico.

Unfortunately no it wouldn’t help much. Reason being is the mushrooms are so dry, so as soon as you start breaking them open or moving them around, then spores (if present) quickly go airborne. One other thing you can do is take a metal straw and take a core sample from the center, but even doing this, we still had contaminated plates here and there.
Yes, soak a razor blade in iso, dry and use that to split the dried fruit and take a sample from the center of fruit that has not been exposed until opening up.

Tecnically this depends on whether or not you know the genetic history. Id you do know the gen # feel free to keep track of it in case theres ever complications that end up arising due to that late gen variable, but if you dont know, starting with gen 1 is how i usually go about it since gen 0 is *tecnically* the dried mushroom you're cloning. But im a novice, im sure theres smarter ways to go about it.
Patreon puts your content at risk. They're well known for censorship. Just because they've ignored you for now, doesn't mean they won't shut you down whenever they feel like it. I suggest transitioning to alternatives (even if you also stay on patreon) Locals is a good option for influencers. It allows you to lock content behind pay walls, or not, however you want to lay it out. They also have their own servers so Amazon can't ever just yeet them from the service. Diversify so your income isn't at such risk. On that note I'd also suggest Odysee as a place to back up your youtube vids.
I'm just curious about the last part. If you're worried about services removing content, then why use cloud services at all for backup? I would think keeping your own copies is the safest way, but what do I know.
come on man you should have showed at least some growth on the agar plate to validate this whole video. Its widely known that doing this method doesn't work. Reviving a dried fruit requieres way more technology than just putting the mushroom in water and throwing it on a plate

Tecnically this depends on whether or not you know the genetic history. Id you do know the gen # feel free to keep track of it in case theres ever complications that end up arising due to that late gen variable, but if you dont know, starting with gen 1 is how i usually go about it since gen 0 is *tecnically* the dried mushroom you're cloning. But im a novice, im sure theres smarter ways to go about it.

I suggest transitioning to alternatives (even if you also stay on patreon) Locals is a good option for influencers. It allows you to lock content behind pay walls, or not, however you want to lay it out. They also have their own servers so Amazon can't ever just yeet them from the service. Diversify so your income isn't at such risk.
