Measuring + Isolating Fluorescence from a Combined Brightfield + Fluorescence image in ImageJ / FIJI

- A viewer contacted me about a situation that seemed a bit different from doing color intensity measurement on a typical color image in ImageJ/FIJI. If you have a combined image where brightfield (or phase contrast or DIC) and a fluorescence channel are merged together, and if you do not have the separate original images/channels, you can do one of two approaches shown in this video to specifically measure the intensity of a selected area's fluorescence. Although the second method is more complex, you will be able to entirely separate the brightfield image from the fluorescence image. This is important as the white & grays from the brightfield image will still contribute green values to any measurements - and thus any green value measurements would not solely be from the fluorescence.
